Azoxymethane and DSS-Induced Colitis Model

C57Bl/6 mice from Harlan Laboratories and the MK2−/−tm1Mgl (8 (link)) mouse strain were bred under pathogen free conditions. Animal procedures were approved by the UNM IACUC. Azoxymethane (AOM), (Sigma Aldrich, St. Louis, MO) was injected IP into 6–8 week old female mice at 12.5mg/kg. Dextran Sodium Sulfate (DSS), (MP Biomedicals, MW 36,000–50,000) was added to drinking water at 2.5% at days 5 and 26 and at 2.0% at day 47, 2.0% for 5 days sacrificed at day 80. Control mice received PBS IP and no DSS in water.

Ultramicronized PEA Characterization and Use

Azoxymethane-Induced Colorectal Cancer Model

Thirty-six BALB/c female mice (6–8 weeks old) were purchased for this animal study (Beijing Vital River Laboratory Animal Technology Co., Ltd.). The study involving the experimental animals was approved by the Institutional Animal Committee of Wenzhou Medical University. The mice were divided into three groups: control group, Fn + NaCl (Beyotime Institute of Biotechnology) group, and Fn + azoxymethane (AOM; 12 mg kg–1; Sigma-Aldrich, United States) group, with 12, 10, and 14 mice in each group, respectively. Each mouse weighed about 20 g. After 1 week of acclimatization, AOM/0.9% NaCl was injected intraperitoneally once. One week later, the mice were given the Fn/0.9% NaCl by oral gavage, once a day, for 5 consecutive days. One week after stopping the intervention, mice were given the intraperitoneal injection of AOM/0.9% NaCl. One week after the second AOM/0.9% NaCl injection, Fn was given via another oral gavage for 5 days. Lastly, the mice were given water and normal food until the end of the experiment (81st day) (Supplementary Figure 1). The concentration of the intraperitoneal AOM injection was 10 mg/kg. The amount of Fn used was 1 × 108 per mouse (OD600 nm = 0.1) with 200 μL 0.9% NaCl.

Colorectal Cancer Induction in T. crassiceps-Infected Mice

An extensively used CAC model was developed as previously described 9 (link). Briefly, T. crassiceps-infected mice (6 weeks post-infection) or age-matched uninfected mice were i.p. injected with Azoxymethane (AOM) at 12.5 mg/kg (Sigma, USA). Five days later, dextran sulfate sodium (DSS, MW: 35 000-50 000, MP Biomedicals. Solon, OH, USA) dissolved at 2% in drinking water was administered ad libitum for 7 consecutive days. Afterwards, the mice were maintained with regular water for 14 days and were subjected to two more DSS cycles. The mice were sacrificed on day 68; the colon was removed, weighed and submitted for macroscopic inspection, histopathological examination and mRNA expression assays. The mouse infection scheme and treatment are showed in Fig. 1A.
