Genetically Modified Mice for Immunology

All mice were on a C57Bl/6 background. Itk−/−, TCRδ (γδ T cell-deficient mice, B6.129P2-Tcrdtm1Mom/J, Jax Labs), IL4Rα−/− (a kind gift from Dr. Frank Brombacher, University of Cape Town, South Africa, via Dr. Fred Finkelman, University of Cincinnati51 (link)). IL17A-GFP reporter mice (C57BL/6-Il17atm1Bcgen/J were purchased from Biocytogen, and Foxp3-RFP reporter mice (C57BL/6-Foxp3tm1Flv/J) were from the Jackson Laboratory (Bar Harbor, ME). Reporter strains were crossed to generate IL17A-GFP/Foxp3-RFP dual reporter strains in WT or Itk−/− background52 (link). TCRδ/Itk−/− were previously described 53 (link). All mice were housed in a specific pathogen-free environment, and male and female mice were between 6 and 8 weeks of age when used. All experiments were approved by the Office of Research Protection’s Institutional Animal Care and Use Committee at The Pennsylvania State University and Cornell University.

Generation of Dual Reporter Mice for Treg and Th17 Cells

CD45.1 (B6.SJL-PtprcaPepcb/BoyJ; 002014), Rag1−/− (B6.129S7-Rag1tm1Mom/J; 002216), IL-10GFP (B6(Cg)-Il10tm1.1Karp/J; 014530)51 (link) and Foxp3RFP (C57BL/6-Foxp3tm1Flv/J; 008374)52 (link) reporter mice were purchased from the Jackson Laboratory (Bar Harbor, ME). IL-17AGFP (B-IL17-EGFP KI; available at Jackson Laboratory as C57BL/6-Il17atm1Bcgen/J; 018472) reporter mice were from Biocytogen (Worcester, MA)53 (link). Reporter strains were crossed to generate IL-10GFP/Foxp3RFP or IL-17AGFP/Foxp3RFP dual reporter strains in WT, Itk−/− or Itkas background. ITKas/Itk−/− (Tg(hCD2-Itkas)Itk−/−) mice, that is, Itkas, were as previously described16 (link)20 (link), and harbour an altered ATP binding pocket (F434G/Δ429) in ITK kinase domain allowing specific inhibition by 1-(tert-Butyl)-3-(3-methylbenzyl)-1H-pyrazolo[3,4-d]pyrimidin-4-amine (3MBPP1). All mice were bred in house, and both female and male mice were used between 5 and 12 weeks of age. All experiments were approved by the Office of Research Protection’s Institutional Animal Care and Use Committee at Cornell University.
