Cell apoptosis was analyzed using Annexin V-FITC/propidium iodide (PI) Double Staining Kit (BD Biosciences, San Jose, California) according to the manufacturer’s instructions. Briefly, transfected cells were washed twice with cold PBS and resuspended in 500 µL binding buffer. Next, cells were stained with 5 µL Annexin V-FITC and 5 µL PI for 15 minutes in dark at room temperature. Then apoptotic cells were analyzed with a flow cytometer equipped with Cell Quest 3.0 software (BD Biosciences).

An Annexin V-FITC/propidium iodide (PI) Double Staining Kit (BD Biosciences, San Jose, CA, USA) was used to detect cellular apoptosis according to the manufacturer's instructions. Cells were collected, washed with cold phosphate-buffered saline, and resuspended at 1 × 106 cells/ml in 1 ml binding buffer. Next, 5 μl Annexin V-FITC and 5 μl PI were added and incubated for 15 min at room temperature in the dark. Cells were detected by flow cytometry. All experiments were repeated three times.

After 72 h of transfection, cells were harvested and fixed in 70% cold ethanol at -20 °C overnight, followed by incubation with FITC-labelled Annexin V and PI for 30 min in the dark. The extent of cell apoptosis, indicated as the percentage of apoptotic cells, was measured using a FACSCalibur flow cytometer (BD Biosciences, Franklin Lakes, NJ, USA). All experiments were performed three times.
