1

Quantifying Serum Hormone Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
Amylin, GLP-1, GIP and Ghrelin (Ray Biotech, USA) levels in serum were tested using ELISA Kit in line with the competitive enzyme immunoassay principle. In brief, 100 μL serum was added to each well and OD values were detected at 450 nm using a spectrophotometer (Synergy2, USA).

Serum Lipid and Adipokine Profiles

Check if the same lab product or an alternative is used in the 5 most similar protocols
The profiles of serum TG, TC, and HDL-C were measured using a clinical chemistry analyzer (Dri-Chem 3500, Fuji Film Co., Tokyo, Japan), while ghrelin (RayBiotech, Inc., Norcross, GA, USA) and leptin (R&D Systems, Minneapolis, MN, USA) levels were analyzed using commercial assay kits, as per the manufacturers’ instructions. The non-HDL-C concentration was calculated as follows: [non-HDL-C]=[TC] – [HDL-C].

Metabolic Biomarkers in Blood

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was obtained from the angular vein with capillary glass tubes for serum analysis. Serum insulin (Crystal Chem, IL, USA), leptin (Crystal Chem), ghrelin (RayBiotech, GA, USA), amylin (RayBiotech), cholesterol (Novus, CO, USA), and triglyceride (Novus) were assayed according to the manufacturer's instructions. Homeostasis model assessment-estimated insulin resistance (HOMA-IR) was calculated as described [21 (link)].

Serum Lipid and Hormone Profiling

Check if the same lab product or an alternative is used in the 5 most similar protocols
Upon completion of the experiments, all rats were weighed and blood was collected from the anesthetized animals into blood collection tubes after a 12-h overnight fast. After standing for 30 min, the serum was prepared by centrifugation of blood at 1000 × g for 10 min at 4 °C and stored at −80 °C until analysis. Serum levels of triglyceride (TG), total cholesterol (TCH), high density lipoprotein-cholesterol (HDL-c), low density lipoprotein-cholesterol (LDL-c), free fatty acid (FFA) and glucose (GLU) were measured by means of enzymatic methods, using assay kits (Sekisui Medic; Abbott Laboratories or Beijing leadman biochemistry co., LTD). Enzyme-linked immunosorbent assays were used to measure the serum levels of leptin (R&D systems, USA), ghrelin (RayBiotech, USA) and adiponectin (CUSABIO Biotech, Ltd).

Gastric Biomarkers Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
The concentrations of gastrin, ghrelin, PGE 2 and histamine in serum as well as in the supernatant of gastric tissue homogenates were measured using commercially available ELISA kits (gastrin, Ray Biotech, Inc., Norcross, GA, USA; and PGE 2 , ghrelin and histamine, Glory Science Co., Ltd, Del Rio, TX, USA). Each assay was performed in duplicate according to the manufacturer's instructions. The minimal detection limits for gastrin, PGE 2 , ghrelin and histamine were 9.92 pg ml -1 , < 0.24 ng l -1 , < 0.24 pg ml -1 and < 0.54 ng ml -1 , respectively.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
