Nasopharyngeal Cancer Cell Lines: Cultivation and Reagents

Human nasopharyngeal cancer cell lines, CNE-1 and CNE-2, purchased from the Chinese Academy of Sciences Cell Bank (Shanghai, China) were maintained in RPMI-1640 medium supplemented with 10% newborn calf serum (NBCS), 100 u/ml penicillin and 100 µg/ml streptomycin at 37˚C in a humidified atmosphere of 5% CO 2 and 95% air. The 293T cell line was obtained from Invitrogen (Carlsbad, CA, uSA) and cultured in Dulbecco's modified Eagle's medium (DMEM) containing 10% fetal bovine serum (FBS; Invitrogen), 100 u/ml penicillin and 100 µg/ml streptomycin. Thiostrepton is a cycling polypeptide antibiotic with selective inhibitory activity against FoxM1 (23) and was purchased from Tocris Cookson (Ellisville, MO, uSA). Antibodies against FoxM1, E-cadherin, zinc finger E-box binding homeobox 2 (ZEB2), zinc finger protein Snail2 and glyceraldehyde 3-phosphate dehydrogenase (GAPDH) were all purchased from Santa Cruz Biotechnology (Santa Cruz, CA, uSA).
