Insulin Secretion in Glucose and IBMX

Cells were preincubated for 30 min in Krebs Ringer bicarbonate buffer (Krebs and Henseleit, 1932) containing 10 mM HEPES pH 7.4, 2 mg/ml bovine serum albumin and 1 mM glucose, and then incubated for 1h in either 1 mM glucose, 20 mM glucose, 1 mM glucose + 0.5 mM IBMX or 20 mM glucose + 0.5 mM IBMX, at 37°C in Krebs Ringer Bicarbonate buffer with the same additions as during the pre-incubation.
