Immortalized human bronchial epithelial cells (HBEC) were obtained from J. Minna (UT Southwestern Medical Center) and were cultured in keratinocyte serum-free media (Invitrogen) supplemented with EGF and bovine pituitary extract. Human embryonic kidney (HEK) 293T cells were cultured in DMEM (Invitrogen) with 10% FBS (Atlas Biologicals). The following antibodies were used for immunoblot analysis according to the manufacturer’s instructions: anti-STING rabbit polyclonal (Cell Signaling), anti-MAVS rabbit polyclonal (Enzo Life Sciences), anti-TRAF3 rabbit polyclonal (Cell Signaling Technology), anti-P65 rabbit polyclonal (Santa Cruz), anti-IRF3 rabbit polyclonal (Santa Cruz), and anti-β-actin mouse monoclonal antibodies (Sigma). Antibodies against VSV proteins were used as previously reported25 (link). The miRIDIAN human microRNA mimic library (miRBase version 10.1) was obtained from Dharmacon, Thermo. A total of 810 mimics were screened, which represented 723 unique mature miRNAs. Non-targeting control Caenorhabditis elegans miRNAs and siRNAs were purchased from Dharmacon. Recombinant human IFNβ was obtained from R&D systems.
