Analytical Technique for Soil Contaminants

The used substrate papers, glass fiber film (SB08, CB06, BT53, RB45), polyester fiber film (VL78), and special absorbent paper (SX18) were purchased from Shanghai Jinbiao Biological Technology Co., Ltd. (Shanghai, China), cadmium oxide, selenium powder, liquid paraffin, oleic acid, oleyl-amine, tetra-methoxy-silane(TMOS), tetra-ethoxy-silane (TEOS), tetra-propoxy-silane (TPOS), 3-aminopropyltriethoxylsilane (APTES), trimethoxy-silyl-propane-thiol (MPTMS), and vinyl-trimethoxy-silane(VMS) were supplied by Aladdin (Shanghai, China); urea, sodium dodecyl sulfonate (SDS), sodium chloride, trichlorfon, acephate, fenthion and dimethoate were supplied by Sigma-Aldrich (Shanghai, China). Soybean samples were purchased from a local market in Xi’an (Shaanxi, China). All the water used in the experiment was ultrapure. All reagents used were of at least analytical grade.

All other chemicals used were analytical grade. Doubly distilled water was used throughout all experiments.

Compound Library Preparation and In Vivo Testing

The FDA Pharmakon compound library was purchased from MicroSource Discovery Systems, Inc. (USA). Compounds (10 mM, dissolved in DMSO) were stored at -80 °C until use. Levamisole and ivermectin, used for the in vitro controls, were purchased from Fluka and Sigma-Aldrich, respectively (Buchs, Switzerland), dissolved at 10 mM stock solutions in DMSO, and stored at -20 °C until further use. For the in vivo studies, bitoscanate, chlorcyclizine HCl, clemastine, dicyclomine HCl, drofenine HCl, ethopropazine HCl, lansoprazole, metformin HCl, morantel, natamycin, trichlorfon, and trimipramine maleate were purchased from Sigma-Aldrich.

HPLC-grade acetonitrile (ACN), methanol, and n-hexane were acquired from Pharmaco-Aaper (Brookfield, CT, USA). All other chemicals and reagents were of analytical grade, unless otherwise specified.

Neurodegeneration Assay in C. elegans

The model organism used in the study was C. elegans N2 (wildtype strain), which was fed upon bacterial food strain i.e., Escherichia coli OP50. Both were received from the Caenorhabditis Genetics Center (CGC, Minneapolis, MN). For neurodegeneration, OH11124 (otIs388 [eat-4(fosmid)::SL2::YFP::H2B + (pBX) pha-1(+)] III ) expressing glutamatergic neuron with YFP expression and OH13083 (otIs576 [unc-17(fosmid) ::GFP + lin-44::YFP]) expressing cholinergic neuron with GFP expression were purchased from CGC. Pesticides used in the study were Chlorpyrifos, Disulfoton and Trichlorfon purchased from Sigma Aldrich. All other chemicals used in the experiments were of analytical grade.
