Chromosomal Tagging of Exocyst Complex

Check if the same lab product or an alternative is used in the 5 most similar protocols
Chromosomal tagging of SEC5, SEC8, EXO70 and EXO84 by Green Fluorescence Protein (GFP) was performed as previously described [38 (link)]. Cells were grown to log phase at 25 °C. The cells were then fixed for microscopy as described previously [39 (link)]. F-actin was stained with Alexa Fluor 488-conjugated phalloidin (Molecular Probes Corp.). Fluorescence microscopy was conducted with microscope CTR6000 (Leica) equipped with a Plan-Apochromat 100x, 1.40 NA oil immersion objective lens. Images were taken using LAS AF 1.5.1 acquisition software (Leica).

Fluorescence Microscopy of Log-Phase Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Fluorescence microscopy was performed with a Leica CTR6000 fluorescence microscope equipped with a Plan-Apochromat 100 × , 1.40 NA oil immersion objective lens. Images were taken using LAS AF 1.5.1 acquisition software (Leica). Cells were grown to early log phase (0.6 OD600) in synthetic complete media and fixed by methanol/acetone before microscopy. Images were captured with a digital camera (DFC350FX; Leica) at room temperature.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
