Cell Culture of Human Ovarian Cell Lines

The human EOC cell lines OVCAR-3, Caov-3, ES-2 and A2780 were purchased from the Chinese Academy of Sciences Cell Bank (Shanghai, China). The human normal ovarian epithelial cell line HOSEpiC were purchased from the American Type Culture Collection (ATCC, USA). OVCAR-3, Caov-3, A2780, and HOSEpiC were cultured in RPMI 1640 medium (Procell, Wuhan, China), supplemented with 10% fetal bovine serum (FBS; Procell). ES-2 was cultured in McCoy’s 5 A medium (Procell), also supplemented with 10% FBS. All cells were maintained under controlled conditions at a temperature of 37°C in an atmosphere containing 5% CO2. Regular monitoring for mycoplasma contamination was performed using PCR.

Ovarian Cancer Cell Line Cultivation

The ovarian cancer cell lines A2780, ES-2, OVCAR-3 and Caov-3 were purchased from Procell Life Science & Technology Co., Ltd. SKOV-3 and 293T cells were obtained from Shanghai Zhongqiaoxinzhou Biotechnology Co., Ltd. A2780, Caov-3 and 293T cells were cultured in DMEM (HyClone; Cytiva) supplemented with 10% FBS (Biological Industries). ES-2 cells were cultured in McCoy's 5A medium (Procell Life Science & Technology Co., Ltd.) supplemented with 10% FBS. OVCAR-3 cells were cultured in RPMI-1640 medium (HyClone; Cytiva) supplemented with 20% FBS and 0.01 mg/ml bovine insulin (Beijing Solarbio Science & Technology Co., Ltd.). SKOV-3 cells were cultured in RPMI-1640 medium supplemented with 15% FBS. All cells were incubated at 37˚C in an incubator with 5% CO2.
