All rodent studies were approved by and performed according to the guidelines of the Institutional Animal Care and Use Committee of the Helmholtz Center Munich, Germany; the University of Cincinnati; or the Augusta University, USA. Wild-type male mice (C57BL/6JRj) were purchased from Janvier (Le Genest-Saint-Isle, France); POMC^eGFP (Stock No: 009593) and NPY^eGFP (Stock No: 006417) male mice were purchased from Jackson Laboratory (Bar Harbor, ME, USA) with the C57BL/6 genetic background. Ten-week-old male Wistar rats were purchased from Charles River (Sulzfeld, Germany). All mice and rats were group housed on a 12 h light, 12 h dark cycle (light from 7:00 to 19:00) at 22 °C, with free access to standard chow diet (LM-485, Teklad) and water before experiments. DIO was induced by HCHF diet purchased from Research Diets (D12331, Research Diets Inc., New Brunswick, NJ, USA). HCHF diet feeding was started at week 8 of age. Body weight and food intake were monitored according to each experimental design. Mice were single-housed after receiving stereotaxic infusions or brain surgeries.

Non-transgenic C57Bl/6 mice were obtained from Charles River (Manston, Kent, UK), Envigo (Huntingdon, UK and Indianapolis IN) or Janvier Labs (Le Genest-Saint-Isle, France). Pomc^eGFP, Cck^ires-Cre, Crh^ires-Cre (Taniguchi et al., 2011 (link)), Slc17a6^ires-Cre and Rosa26-loxSTOPlox-eYFP were all purchased from Jackson Laboratories (stock numbers 009593, 012706, 012704, 016963 and 006148, respectively; Bar Harbor, ME). We have described the generation of the PPG (Gcg^Cre) and PrRP (Prlh^ires-Cre) mice (Parker et al., 2012 (link); Dodd et al., 2014 (link)). Calca^ires-Cre mice (Carter et al., 2013 (link)) were a kind gift from Prof Richard Palmiter (Howard Hughes Medical Institute, University of Washington).

All mouse care and experimental procedures were approved by the Institutional Animal Care Research Advisory Committee of the Albert Einstein College of Medicine. Mice used in these experiments included POMC-Cre (stock # 005965), POMC-eGFP (stock # 009593), TRPV1-Cre (stock # 017769), floxed-stop CRISPR/Cas-9-eGFP mice (stock # 024857), and TRPV1-KO (stock # 003770, the Jackson Laboratory). Both male and female mice of mixed C57BL/6, FVB, and 129 strain backgrounds were used.

All mice were housed in a pathogen-free animal facility under a normal 12 h light, 12 h dark cycle with ad libitum access to normal chow and water, unless otherwise noted. All studies were approved by the Institutional Animal Care and Use Committee of Oregon Health and Science University. Npy-hrGfp (#006417), Pomc-eGfp (#009593) and Nkx2.-Cre (#008661) mice were purchased from the Jackson Laboratory. Otp^+/− and Dlx1/2^f/f mice were kindly provided by Dr. Dario Acampora and Dr. Magdalena Petryniak, respectively.

All animals were housed under a 12/12 h light/dark cycle at a temperature of 20 ℃ to 22 ℃. Standard pellet chow and water were provided ad libitum. The animal care and procedures were approved by the Animal Core Facility of Nanjing Medical University. NPY-hrGFP (Jax#006417), and POMC-EGFP (Jax#009593) mice were purchased from the Jackson Laboratory (Bar Harbor, ME, USA). The Cre-dependent Rosa26 Cas9 knockin mice were kindly provided by Professor Bin Shen (Nanjing Medical University, China).
