Transgenic Mouse Model for Neuroendocrine Studies

A total of n=32 (18 male and 14 female) transgenic C57BL/6-Tg(Pomc-EGFP)1Low/J (Pomc-EGFP, Jackson laboratories, stock #009593) mice, ages 14–32 weeks, were used for these experiments (Cowley, et al. 2001 (link); Overstreet, et al. 2004 (link)). Mice were housed in a temperature controlled vivarium (22–24°C) on a 14hr/10hr light/dark cycle (lights off at 7 am). A non-standard light cycle was chosen based on Jackson Laboratories breeding colony recommendations. Mice were given standard rodent chow and water ad libitum. All experiments were conducted under Arizona State University Institutional Animal Care and Use Committee approval, and were conducted during the dark phase of the light cycle.

POMC, AgRP, and CRISPR Mouse Models

All experimental and animal care protocols were approved by the Gyeongsang National University Institution Animal Care and Use Committee (GNU IACUC, GNU-170504-M0019) and performed in accordance with the National Institutes of Health (NIH) guidelines and with a scientifically reviewed protocol (GLA-100917-M0093). Mice used in these experiments included POMC-Cre (stock # 005965, Jackson Laboratory, Bar Harbor, ME, USA) and POMC-eGFP (stock # 009593, Jackson Laboratory, Bar Harbor, ME, USA), AgRP-IRES-Cre (stock # 012899, Jackson Laboratory, Bar Harbor, ME, USA), CRISPR/Cas9 (stock # 026175, Jackson Laboratory, Bar Harbor, ME, USA), which are mixed C57BL/6, FVB, and 129 strain backgrounds. CRISPR/Cas9 mice were used for the Cre recombinase-dependent expression of eGFP.

Murine Phenotyping: Genetic Knockout

8–14 week-old male and female C57BL/6J wild type (WT, JAX catalog number # 000664), Lipocalin 2 knockout (Lcn2-KO, JAX catalog number # 024630), and Pomc-EGFP (JAX catalog number # 009593) mice were purchased from The Jackson Laboratory (Bar Harbor, ME) and maintained in our animal facility. Sex of mice used in each study is specified in corresponding figure legends. Briefly, Lcn2-KO mice were generated on a C57BL/6J background. All mice were housed and bred in a dedicated mouse room with a temperature 26°C with a 12-hour light/dark cycle. Animals were provided ad libitum access to food and water (Rodent Diet 5001; Purina Mills). Lcn2-KO mice were genotyped according to the standard protocol from The Jackson Laboratory. In behavioral studies, animals were individually housed for acclimation at least 7 days prior to interventional studies. Mouse studies were conducted in accordance with the National Institutes of Health Guide for the Care and Use of Laboratory animals, and approved by the Institutional Animal Care and Use Committee (IACUC) of Oregon Health & Science University.

Mouse and Rat Models of Obesity

All rodent studies were approved by and performed according to the guidelines of the Institutional Animal Care and Use Committee of the Helmholtz Center Munich, Germany; the University of Cincinnati; or the Augusta University, USA. Wild-type male mice (C57BL/6JRj) were purchased from Janvier (Le Genest-Saint-Isle, France); POMCeGFP (Stock No: 009593) and NPYeGFP (Stock No: 006417) male mice were purchased from Jackson Laboratory (Bar Harbor, ME, USA) with the C57BL/6 genetic background. Ten-week-old male Wistar rats were purchased from Charles River (Sulzfeld, Germany). All mice and rats were group housed on a 12 h light, 12 h dark cycle (light from 7:00 to 19:00) at 22 °C, with free access to standard chow diet (LM-485, Teklad) and water before experiments. DIO was induced by HCHF diet purchased from Research Diets (D12331, Research Diets Inc., New Brunswick, NJ, USA). HCHF diet feeding was started at week 8 of age. Body weight and food intake were monitored according to each experimental design. Mice were single-housed after receiving stereotaxic infusions or brain surgeries.

Genetically Modified Mouse Strains

Non-transgenic C57Bl/6 mice were obtained from Charles River (Manston, Kent, UK), Envigo (Huntingdon, UK and Indianapolis IN) or Janvier Labs (Le Genest-Saint-Isle, France). PomceGFP, Cckires-Cre, Crhires-Cre (Taniguchi et al., 2011 (link)), Slc17a6ires-Cre and Rosa26-loxSTOPlox-eYFP were all purchased from Jackson Laboratories (stock numbers 009593, 012706, 012704, 016963 and 006148, respectively; Bar Harbor, ME). We have described the generation of the PPG (GcgCre) and PrRP (Prlhires-Cre) mice (Parker et al., 2012 (link); Dodd et al., 2014 (link)). Calcaires-Cre mice (Carter et al., 2013 (link)) were a kind gift from Prof Richard Palmiter (Howard Hughes Medical Institute, University of Washington).

Genetic Mouse Models for Neuroscience

Conditional Knockout of TTF-1 in Mice

All animals were housed individually in a temperature-controlled room with a 12-hour light–dark cycle and had ad libitum access to a standard chow diet (Feedlab, Gyeonggi-Do, Korea) and water unless otherwise stated. Transgenic mice (POMC-EGFP [stock no. 009593], ObRb-Cre [stock no. 008320], POMC-Cre [stock no. 005965] and Ai14-Cre [stock no. 007914]) were obtained from Jackson Laboratory (Bar Harbor, ME, USA). TTF-1f/f mice [15 (link)] were crossbred with ObRb-Cre or POMC-Cre mice to generate TTF-1 conditional knockout mice specifically in the cells expressing ObRb or POMC. The resulting mouse lines that bore the TTF-1 deletion in the POMC cells (POMC-TTF-1−/−) and in the cells expressing ObRb (ObRb-TTF-1−/−) were first subjected to IHC observations to identify the specific deletion of TTF-1 in each cell (Supplementary Figures 2 and 3). All animals and procedures used were in accordance with the guidelines and approval of the Institutional Animal Care and Use Committee at the University of Ulsan (permission numbers: BJL-14-050, BJL-14-052 and BJL-17-030).

Mouse Housing and Transgenic Breeding

Mouse Husbandry and Genotyping

All NPY-hrGFP, POMC-EGFP, and C57BL/6J mice were purchased from The Jackson Laboratory. The transgenic mice were genotyped for Gfp. Only male mice were used. All the mice were kept in temperature-and humidity-controlled rooms on a 12-hr:12-hr light-dark cycle, with lights on from 7:00 a.m. to 7:00 p.m. Mouse chow (PicoLab Rodent Diet 20, 5053 tablet, TestDiet) and water were provided ad libitum unless otherwise noted. The mice were single-caged once they received viral injections or were inserted with the guide cannula until all the experiments on the mice were finished.
