Iododeoxyuridine

Manufactured by Merck Group

Iododeoxyuridine is a synthetic nucleoside analog used as a molecular biology research tool. It can be incorporated into DNA in place of thymidine, and is commonly used to label and track proliferating cells.

DNA Replication Inhibitor Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Camptothecin (Sigma-Aldrich) was dissolved in DMSO and a stock solution (10 mM) was prepared and stored at −20°C. Iododeoxyuridine (IdU) and chlorodeoxyuridine (CldU) (Sigma-Aldrich) were dissolved in sterile DMEM to obtain stock solutions of 2.5 mM and 200 mM, respectively, and stored at −20°C. Mirin (Calbiochem), an inhibitor of MRE11 exonuclease activity, was used at 50 μM; the B02 compound (Selleck), an inhibitor of RAD51 activity, was used at 27 mM. The CDK inhibitor roscovitine (Selleck) was used at 20 μM.

Cell Cycle Dynamics in TUBG1 Mutant U2OS Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
U2OS, TUBG1-sgRNA-U2OS-TUBG1, and TUBG1-sgRNA-U2OS-TUBG1A429-A432-A435 cells were seeded at a density of 1.5 × 105 cells into 6-well plates. The cells were treated with 6 μM aphidicolin (Biochemica, #10797) for 24 h, followed by pulse-labeling with 25 μM chloro deoxyuridine (Sigma-Aldrich, C6891) for 30 min and sequentially with 250 μM iodo-deoxyuridine (Sigma-Aldrich, I7125) for 30 min. Collected cells were diluted to 5 × 104 cells/mL and then spread and stained as previously described52 (link). Images were acquired with a Zeiss LSM 780 inverted confocal microscope and analyzed with the ImageJ software.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
