Colistin

Manufactured by Solarbio
Sourced in China

Colistin is a bactericidal antibiotic used in laboratory settings to inhibit the growth of certain Gram-negative bacteria. It acts by disrupting the bacterial cell membrane, leading to cell death. Colistin is commonly used in microbiological research and antimicrobial susceptibility testing.

CRKP Antimicrobial Susceptibility Testing

Check if the same lab product or an alternative is used in the 5 most similar protocols
The antimicrobial susceptibility testing of the CRKP from HWW, recipient and transconjugants was performed by Vitek-2 (Vitek-AST-GN16) (BioMerieux, France) according to the Clinical and Laboratory Standards Institute guidelines (CLSI, 2022). The antibiotics included ampicillin, amoxicillin/clavulanic acid, piperacillin/tazobactam, cefazolin, cefoxitin, ceftriaxone, cefepime, aztreonam, amikacin, gentamicin, tobramycin, ciprofloxacin, levofloxacin, nitrofurantoin, and trimethoprim/sulfamethoxazole. Among them, MICs of ertapenem (Macklin, China), imipenem (Solarbio, China), meropenem (Solarbio, China), tigecycline (Macklin, China) and colistin (Solarbio, China) were determined using broth dilution method. E. coli ATCC 25,922 was used as quality control.

Antimicrobial Susceptibility Testing of MNPECs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Antimicrobial susceptibility testing (AST) was performed by broth microdilution method as our previously described (Liu et al., 2021b (link)). Briefly, 0.5 McFarland inoculum suspensions were diluted 1:100 to a final inoculum density of 106 CFU/ml. Ampicillin (AMP), ceftazidime (CAZ), cefotaxime (CTX), aztreonam (AZM), tetracycline (TET), doxycycline (DOX), ciprofloxacin (CIP), enrofloxacin (ENR), gentamicin (GEN), sulfamethoxazole (SMZ), fosfomycin (FOS), florfenicol (FFC), colistin (COL), and meropenem (MEM) (Solarbio, Beijing, China) were selected to determine the minimum inhibitory concentrations (MICs) of MNPECs. The MICs were recorded as the lowest concentrations of antimicrobials in the wells, where no visible bacteria growth was observed. Escherichia coli ATCC 25922 was used as standard reference strains.

Synthesized AMPs such as Iseganan (IB), Melittin (GQ) and Cathelicidin-DM (SA) were procured from Dangang Biotechnology Company. Polymyxin B (PB) and colistin (CST) were acquired from Solarbio. Specific information about these five AMPs are shown in Table 1.

Antimicrobial Susceptibility Testing Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Antimicrobial susceptibility testing was performed by broth microdilution method, as internationally recognized clinical guidelines those provided by the Clinical and Laboratory Standards Institute (CLSI) (CLSI, 2013 , 2019 ). Briefly, 0.5 McFarland inoculum suspensions were diluted 1:100 to a final inoculum density of 105 CFU/ml. Ampicillin (AMP), ceftazidime (CAZ), cefotaxime (CTX), aztreonam (AZM), tetracycline (TET), doxycycline (DOX), ciprofloxacin (CIP), enrofloxacin (ENR), gentamicin (GEN), sulfamethoxazole (SMZ), fosfomycin (FOS), florfenicol (FFC), colistin (COL), and meropenem (MEM) (Solarbio, Beijing, China) were selected to determine the minimum inhibitory concentrations (MICs) of MPECs. The antimicrobials were twofold serially diluted ranging from 0.25 to 512 μg/ml and incubated at 37°C for 18 h. The MICs were recorded as the lowest concentrations of antimicrobials in the wells, where no visible bacteria growth was observed.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
