Opioid Metabolite Monitoring in Wastewater

Five parent opioids and their respective metabolites were monitored in raw wastewater. The investigated opioids were morphine (MOR), its major metabolite morphine-3-glucuronide (M3G), codeine (COD), its major metabolite norcodeine (NCOD), oxycodone (OXY), its major metabolite noroxycodone (NOXY), fentanyl (FENT), its major metabolite norfentanyl (NFENT), heroin (HER), and its minor but exclusive metabolite 6-acetylmorphine (6-AM). High purity (>97%) standard solutions of the target compounds originated from Sigma Aldrich (Milwaukee, WI) and were prepared by Cerilliant (Round Rock, TX, USA) as solutions in methanol or acetonitrile. Five deuterated compounds, one for each of the parent opioid target compounds were also purchased from Cerilliant for use as internal standards for quantification: heroin-d9 (HER-d9), morphine-d6 (MOR-d6), codeine-d6 (COD-d6), oxycodone-d3 (OXY-d3), and fentanyl-d5 (FENT-d5).

Comprehensive Fentanyl Analyte Analysis

The primary reference materials for fentanyl, norfentanyl, acetyl fentanyl, acetyl norfentanyl, and their respective internal standards (ISTD) fentanyl-d5, norfentanyl-d5, acetyl fentanyl-13C6, and acetyl norfentanyl-13C6 were purchased from Cerilliant (Round Rock, Texas). Ammonium formate, ammonium hydroxide, glacial acetic acid, isopropanol, methylene chloride, methanol, sodium phosphate monobasic, sodium phosphate dibasic and deionized (DI) water were purchased from Fisher Scientific (Hanover Park, Illinois). All reagents were ACS grade or better. SPEC® MP3 solid phase extraction (SPE) columns were purchased from Agilent Technologies, Inc, (Santa Clara, California). Medical grade nitrogen was purchased from National Welders Supply Company (Richmond, Virginia). Drug-free expired whole blood was obtained from Transfusion Medicine Laboratory at Virginia Commonwealth University Health System. The blood was certified drug free by screening for alkaline extractable drugs by GC/MS and UPLC/MS/MS prior to use.

Methoxyacetylfentanyl Quantification and Metabolites

methoxyacetylfentanyl was obtained from Cayman Chemical (Ann Arbor, MI), and 4-ANPP was obtained from Toronto Research Chemicals (North York, Canada). The internal standard fentanyl-d5 was obtained from Cerilliant (Round Rock, TX). For LC–MS-MS quantification, LC–MS grade acetonitrile, formic acid and methanol were obtained from Fisher Scientific (Gothenburg, Sweden) and ammonium formate from Fluka (Sigma‐Aldrich, Stockholm, Sweden). For sample preparation, gradient grade acetonitrile, methanol and formic acid p.a. (98%) from Merck (Darmstadt, Germany), 95% ethanol from Kemetyl (Haninge, Sweden) and β‐glucuronidase/arylsulfatase (Helix pomatia) from Roche (Mannheim, Germany) were used.

Deionized (DI) water was prepared using a water purification system (18.4 MΩ·cm Facility Scale Reverse Osmosis/Ion Exchange Water Purification System; Applied Membranes Inc, Vista, CA, USA). All standards were prepared gravimetrically, with w/w concentrations represented as ng of analyte per g of total mass.

Quantitative Analysis of Opioids in Urine

For flow-injection analysis, all working stock solutions were diluted to a concentration of 500 ng/mL in LC–MS grade water.

Sensitive Bioanalytical Quantification Method
