The following mouse monoclonal antibodies were used; anti-Tau-1 (MAB3420; Chemicon International, Temecula, CA), anti-Myc 9E10 and anti-MAP2 (M4403; Sigma-Aldrich, St Louis, MO). Polyclonal rabbit antibodies used were anti-GFP (#598; MBL, Nagoya, Japan), anti-RFP (#600-401-379; Rockland Immunochemicals, Gilbertsville, PA), anti-Rbfox2 (Fox2) (A300-864A-T, Bethyl Laboratories, Montgomery, TX) and anti-Sept1129 (link). anti-GFP (GFP-1020; Chicken) was purchased from AVES Labs (Tigard, OR).

Immunostaining of Neuronal Cultures

Cultures were fixed (4% PFA, 20 min RT) and immunostained as described in [13 (link)]. Primary antibodies included anti-α-tubulin-FITC (F2168, sigma), anti-MAP2 (M4403, Sigma), anti-Synaptophysin (S5768, Sigma), anti-Vglut1, anti-α-synuclein (4179, cell signaling), anti-pTau Thr231 (44746G, Invitrogen). Species-specific secondary antibodies (coupled to Alexa 350, 488, 555) were used (Invitrogen). Images were acquired with an Axio-observer Z1 (Zeiss) fitted with a cooled CCD camera (CoolsnapHQ2, Ropert Scientific) and images were analyzed using ImageJ.

Following fixation with 4% paraformaldehyde, cells was incubated in blocking serum (10% fetal bovine serum in 0.2% PBST) for 1 h. Primary antibodies include M4M (0.4 µg/ml), anti-Myc (C3956, Sigma-Aldrich, MI, USA), anti-WGA (Cat#W11261, Thermo Fisher Scientific, MA, USA), IgG from human serum (I4506, Sigma-Aldrich, MI, USA), anti-MAP2 (M4403, Sigma-Aldrich, MI, USA). Secondary antibodies are conjugated with FITC or Alexa Fluor 594, Alexa Fluor 647.
