HL-60 human peripheral blood lymphoblast cells were obtained from the American Type Culture Collection (USA) and cultured in RPMI-1640 medium containing 10% FBS and 1% penicillin-streptomycin. The cells were seeded into 96-well black plates at a density of 1 × 105 cells/well and incubated for 30 min. After incubation, the cells were co-treated with 100 nM Phorbol 12-myristate 13-acetate (PMA; Sigma Aldrich, USA) and 200 μg/ml LS803 at 37°C for 3 h with 5% CO2. SYTOX Green dye (Invitrogen, USA) was diluted 500-fold with Hank’s balanced salt solution (Welgene) and added to the wells, followed by incubation at 25°C for 10 min. The fluorescence intensity was analyzed at 480/530 nm using a SpectraMax i3 (Molecular Devices, USA).
