Cell-free supernatants were harvested and analyzed for levels of TNFα, IL-6, and IL-12p40 (BD Biosciences, Heidelberg, Germany) by ELISA according to the manufacturer's instructions. Multiplex Cytokine Profiling in Liver Transplantation

A total of 50 chemokines and growth factors in sera of patients who underwent liver transplantation were quantified by Luminex-based multiplex assay according to the manufacturer’s instruction (Bio-Rad, USA). The detection range of all cytokines was between 0.1 pg/mL and 40 µg/mL. Only data points where ‘pre-OLT’ and ‘post-OLT’ measurements for any given patient and mediator were technically successful and within the dynamic range of the assay were included in the analysis.

Simultaneous Evaluation of LDL, HDL, and Serum Proteins

On a 96-wells plate, LDL, HDL and whole serum were measured simultaneously. Protein concentrations in EV fractions and serum were measured in a well-established protein panel consisting of: CD14, cystatin C, serpin C1 and serpin G1 [20 (link)]. Capture antibody, biotinylated detection antibody and antigen of all four proteins were purchased from R&D systems. Proteins were quantitatively analyzed by Luminex-based multiplex assay (Bio-Rad, Austin, USA). All EV protein levels were corrected for total amount of protein.
