Total internal reflection fluorescence (TIRF) microscopy imaging was performed on an Olympus IX83 motorized TIRF microscopy with a 150×/1.45 NA objective and an Andor IXON ULTRA 512X512 EMCCD camera. For each cell the mean intensity of CD3 and ICAM-1 was calculated by dividing the total fluorescence intensity of the region of interest (ROI) minus background fluorescence of a neighbouring area (measured using the same ROI) and then divided by the area of the ROI.

Spinning Disk Confocal Microscopy Protocol

All cells were imaged on a spinning disk confocal microscope except for Figure 3, which was on a Zeiss LSM 880 Confocal Laser Scanning Microscope equipped with a Plan-Apochromat 63× oil objective (1.4 N.A) and Airyscan detector and controlled by Zen Black (Zeiss). The spinning disk is a Nikon Ti2E body with PSF; CSU-X1 Yokogawa confocal scanner unit; OBIS 405, 488, 561, and 640 lasers and an ANDOR iXON Ultra 512X512 EMCCD camera. Images were acquired with Nikon Plan Apo λ 100× oil objective (1.4 N.A). Images were acquired with micro-manager 2.0. Images were processed in Fiji (NIH) and Adobe Illustrator (Adobe).
