Phytochemical Standards and Analytical Reagents

Supercritical CO2 Extraction of Oregano

O. vulgare spp. hirtum aerial parts were commercial samples purchased in the supermarket Diellas Markets (Corfu, Greece). The mean particle size (0.26 mm) of the material was determined using vibration sieve sets (CISA, Cedaceria, Spain). The carbon dioxide (Messer, Novi Sad, Serbia) with purity >99.98% (w/w) was used for the lab-scale scCO2 extraction. Viscozyme, a cellulolytic enzyme mixture (which contains a wide range of carbohydrases, including arabanase, cellulase, β-glucanase, hemicellulase, and xylanase), methanol, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) were purchased from Sigma-Aldrich.

Viscozyme-Assisted Metabolite Extraction

One leaf disc (Ø3 cm) was ground in an Eppendorf tube and mixed with 0.6 mL 50 mM sodium acetate, pH 5.3. To this, 20 µL of Viscozyme (Sigma-Aldrich) or water (control) was added prior to shaking for 1 h at room temperature. The Viscozyme treated sample appeared viscous compared to control. Metabolites were extracted in 1 mL ethyl acetate by shaking for 5 min and the upper layer was transferred to new vials. The solvent was evaporated under a stream of nitrogen and the residue re-suspended in 1 mL methanol and analyzed by LC–ESI–MS.

Enzymatic Hydrolysis of Pretreated Lignocellulosic Biomass

Pretreated Corn Stover (PCS) or Pretreated Sugarcane Straw (PSCS) were prepared by steam explosion with diluted sulfuric acid at the Abengoa Bioenergy Biomass Pilot Plant in York (Iowa, USA), following the procedure described by Alcántara et al. (2016 (link)). Hydrolysis of PCS or PSCS (10 g l−1) was performed in 100 mL borosilicate conical flasks containing 1xM9 with pH adjusted to 5.5. The enzymatic cocktail Viscozyme (Sigma‐Merck) was added as indicated in Table S3. Glucan content had been previously determined according to the standard biomass analytical procedures by NREL (Rocha‐Martín et al., 2018 ). Samples were incubated in an orbital incubator at 50 °C with shaking at 70 rpm for 24 h.
