Quantification of Inflammatory Cytokines

Concentrations of TNF-Ɑ, IL-6 and IL-8 were measured in 25 μL of EDTA plasma using an ultrasensitive electrochemiluminescence immunoassay, according to the manufacturer’s instructions (Meso Scale Discovery: MSD; Rockville, MD). Ultrasensitive 10-plex plates were custom-designed for the Meso Scale Discovery 6000 instrument. Controls for standard curves were included in each plate. Samples with cytokine values less than the limit of detection were assigned a value of one half the limit of detection.

Serum IL-8 Quantification by ECLIA

Serum IL-8 levels were measured as previously described (30 (link)). Briefly, blood was collected at the time of enrollment into the case control study and allowed to clot. Serum was isolated by centrifugation and stored at −80°C. After thawing, 25 μL of serum were measured for IL-8 concentrations using electrochemiluminescence immunoassay (ECLIA) plates (MesoScale Discovery, Gaithersburg, MD) and analyzed on the MesoScale Discovery 6000 instrument, following the manufacturer’s assay and analysis protocols. Samples were blinded and randomly distributed for order of processing.
