MCF10A breast epithelial cells were purchased from ATCC and used within 10 passages. Human mammary epithelial cells (HuMEC) were purchased from ThermoFisher Scientific and passage 4 cells used for all experiments. Cas9-expressing MCF10A (MCF10A-Cas9) were generated by transduction with lentiCas9-blast lentivirus (Addgene) and selected with 10 μg/ml blasticidin (ThermoFisher Scientific) 72 hours post-transduction. Cells were cultured at 37 ˚C and 5% CO2 in MCF10A media composed of DMEM/F12 (ThermoFisher Scientific) containing 10% fetal bovine serum (ThermoFisher Scientific), 1% penn-strep (ThermoFisher Scientific), 10 ng/ml EGF (LC Labs), 5 μg/ml insulin (ThermoFisher Scientific), 10 ng/ml cholera toxin (List Labs) and 1 μg/ml hydrocortisone (Sigma).
