Maxillae from TRPV4+/+ and TRPV4−/− mice (n = 3 per group) were fixed with 2.5% glutaraldehyde in 0.1 mol/L sodium cacodylate buffer overnight and decalcified with 10% EDTA. The gingival tissues were carefully separated from the teeth and postfixed with 1% osmium tetroxide for 1 hour at 4°C.
