Characterization of Pesticide Standards

Chlorantraniliprole (98% purity, CAS 500008‐45‐7) and flubendiamide (98.1%, CAS 272451‐65‐7) were obtained in‐house at Bayer AG. Cyprodinil (99%, CAS 121552‐61‐2), iprodione (99.6%, CAS 1215631‐57‐4), boscalid (99.5%, CAS 188425‐85‐6), pyraclostrobin (99.9%, CAS 175013‐18‐0), chlorothalonil (99%, CAS 1897‐45‐6), difenoconazole (99.5%, CAS 119446‐68‐3), fenbuconazole (99%, CAS 114369‐43‐6), metconazole (98.9%, CAS 125116‐23‐6) and propiconazole (99%, CAS 60207‐90‐1) were purchased from Sigma‐Aldrich (St. Louis, MO, USA). The fluorescent probe 7‐benzyloxymethoxy‐4‐(trifluoromethyl)‐coumarin (BOMFC; CAS 277309‐33‐8) was synthesized by Enamine Ltd. (Riga, Latvia) with a purity of 95%. HPLC gradient grade acetonitrile was purchased from Merck (Darmstadt, Germany). Unless otherwise mentioned all other reagents were of analytical grade and obtained from Sigma‐Aldrich (St. Louis, MO, USA).

HNE Impact on Cellular Metabolism and DNA Damage

The 4-hydroxynonenal (HNE) was provided by Clinisciences (Nanterre, France). Fetal Bovine Serum (FBS) provided by ThermoFisher (Eindhoven, Netherlands). Puromycin, hydromycin and zeocin were provided by Fisher Scientific(Hampton, NH, USA). Epidermal growth factor (EGF), hydrocortisone, insulin, transferrin, sodium selenite (5 nM) and gentamycin sulfate (5 µg/mL), DMSO (dimethylsulfoxyde), procymidone, iprodione, cyprodinil, fludioxonil, lambda-cyhalothrin, methyl methanesulfonate (MMS), Trifluoromethoxy carbonylcyanide phenylhydrazone (FCCP), rotenone, oligomycin and etoposide were provided by Sigma-Aldrich (Saint-Quentin Fallavier, France). Ro 19-8022 was a gift from Hoffman Laroche Ltd. (Basel, Switzerland), and formamidopyrimidine-DNA glycosylase (Fpg) a gift from Serge Boiteux, CNRS, France.

Multianalyte Plasma Quantification Protocol

reference standards of diazepam, lorazepam, alprazolam, methaqualone,
omeprazole, acetochlor, atrazine, metolachlor, cyprodinil, and chlorpyrifos-methyl
were purchased from Sigma-Aldrich (St. Louis, MO, USA). The physicochemical
properties of the analytes and the rationale behind the selection
of the targeted analytes are listed in Table S1. diazepam-D5, obtained from Cerilliant-Sigma-Aldrich
(Austin, MO, USA), and metolachlor-D6, purchased from Toronto
Research Chemicals (North York, Ontario, Canada), were used as deuterated
internal standards. Pooled bovine plasma, rabbit plasma, rat plasma,
and human plasma preserved in sodium citrate were purchased from Innovative
Research (MI, USA). The authors are not aware of any ethical issue
associated with plasma collection; Innovative research is ISO, Food
and Drug Administration (FDA), USDA, and EPA approved. C18/PAN SPME fibers were provided by Millipore Sigma (Bellefonte, PA,
USA). The thickness and coating length of the fiber were 45 μm
and 1.5 cm, respectively. LC-MS grade methanol, water, ammonium formate,
and formic acid were obtained from Fisher Scientific (Waltham, MA,
USA), and phosphate-buffered saline (PBS) was purchased from Sigma-Aldrich
(St. Louis, MO, USA).

The pesticide standards that included acetamiprid, boscalid, cyprodinil, dimethoate, dithianon, fenhexamid, fenoxycarb, fenpyrazamine, fludioxonil, fluopyram, iprodione, pyraclostrobin, pirimicarb, tebuconazole, thiacloprid, and trifloxystrobin with a purity in the range of 99.4–100 % were purchased from Sigma Aldrich (Germany). The chemical structures selected compounds are shown in Figures S13-S14. Stock solutions were prepared in acetonitrile and stored at −20 °C. Deionized water for the preparation of the mobile phase was produced by Watek system (Germany). Ammonium formate for mass spectrometry was obtained from Fluka (Germany). Formic acid used in the mobile phase and acetonitrile in HPLC grade was obtained from Sigma-Aldrich (Germany). Methanol (HPLC grade) was from Merck (Germany).

Phenanthrene-d10, pyrene-d10, and a polycyclic aromatic hydrocarbon mix (CRM 47930) were purchased from Sigma-Aldrich.
