Preparation and Characterization of Mix-MA Monomer Benchmark

The blend “Mix-MA”, used as
a benchmark monomer, has been prepared with 33.3 wt % of (hydroxypropyl)methacrylate,
33.3 wt % of 1,4-butanediol dimethacrylate, and 33.3 wt % of a urethane
dimethacrylate monomer, obtained from Sigma Aldrich and represented
in Scheme 2. Mix-MA
was already used in previous works.3 (link),4 (link) More particularly,
its viscosity is well adapted for the preparation of formulations.
In addition, interestingly, Mix-MA is not too viscous. Therefore,
oxygen inhibition occurs, and only good-performance systems can polymerize
it. BlocBuilder-MA was obtained from Arkema. tert-Butylbenzene was obtained from Sigma-Aldrich.

Synthesis of Carboxylated Nanodiamond Composites

The monomers, butyl acrylate (BA; 99%, Sigma-Aldrich, Helsinki, Finland) and N,N-dimethylaminoethyl methacrylate (DMAEMA; 99%, Acros Organics/VWR, Vantaa, Finland), as well as the cross-linker, 1,4-Butanediol dimethacrylate (BuDMA; 95%, Sigma-Aldrich, Helsinki, Finland), were distilled under reduced pressure, and were run through an anhydrous alumina column and a filter to remove the inhibitors and oligomers. The photoinitiator, 2-hydroxy-2-methylpropiophenone (97%, Sigma-Aldrich, Helsinki, Finland), carboxylated detonation nanodiamond powder (μDiamond® Molto Vox; Carbodeon Oy, Vantaa, Finland), and acetonitrile (ACN; 99.9%, Sigma-Aldrich, Helsinki, Finland) were used as received. Deionized ultrapure water with ~18 mΩ/cm resistivity was used as received from the purifier. Carbonate buffer (AVS Titrinorm; VWR, Helsinki, Finland) was used as received.

Antibacterial Hydrogel for Bone Regeneration

PEGMEM [poly(ethylene glycol) methyl ether methacrylate] with average Mn = 300, DEM [2-(dimethylamino)ethyl methacrylate], 1,4-butanediol dimethacrylate, ammonium persulfate (APS), ZnCl2, SrCl2, glutaraldehyde, acetic acid, Thiazolyl Blue Tetrazolium Bromide (MTT), and Alizarin Red S were purchased from Sigma-Aldrich. α-Tricalcium-phosphate (α-TCP) was purchased from Fluka.
Simulated body fluid (SBF) solution was prepared according to British Standard ISO 23317:200.
Gram-positive bacteria, methicillin-resistant Staphylococcus aureus (MRSA)—MRSA (NCTC 12493) and Staphylococcus epidermidis (RP62a), were used. Bacteria frozen stocks were stored at −80°C; weekly, strains were streaked out on Brain Heart Infusion (BHI) plates (Oxoid) and incubated for 24 h at 37°C and then stored at 4°C.
