Myoblast Differentiation Imaging Protocol

Myoblast differentiation was examined according to previously published protocols [18 (link)], and samples were imaged on INCell Analyzer 6000 (GE Healthcare), equipped with 10×/0.45 NA Plan Apo objective and 2048 × 2048 sCMOS camera. Twenty-five fields with five z-planes were acquired. Individual focal plane image for each z-series was automatically selected based on sharpness. Image analysis algorithm and PCR is described in Supplementary Methods.

Automated Imaging of Tau Aggregates in HEK293T Cells

HEK293T cells treated with virus expressing either tau RD S320F, S320F_I328S, or P301S were plated at 10,000 cells per well in media (10% FBS, 1% Pen/Strep, 1% GlutaMax in Dulbecco’s modified Eagle’s medium) in a 96-well clear bottom plate (Corning, Product # 3603). After 24 h, cells were stained with Hoechst33342 and Wheat Germ Agglutinin at a final concentration of 5 μg/mL in media for 10 min at 37 °C, protected from light. The staining solution was removed and substituted with 1x PBS afterward. The plate was placed on an IN Cell 6000 Analyzer (GE Healthcare) with a heated stage, and 50 fields of view were imaged under 4′,6-diamidino-2-phenylindole (DAPI), and FITC channels at ×60 magnification (Nikon ×60/0.95, Plan Apo, Corr Collar 0.11–0.23, CFI/60 lambda). Images were exported as TIFF files for downstream analysis.
