For anti-CD47 antibody injection, mice were injected i.t. with 50–70 µg anti-CD47 (clone MIAP301, BioXCell) 9 or 10 d and 13 or 14 d after tumor inoculation. For anti-CD47 antibody systemic injection, mice were injected i.p. with 500 µg anti-CD47 9 d and 13 d after tumor inoculation. For anti-IFNαR antibody injection, mice were injected i.t. with 200 µg anti-IFNαR antibody 6, 8, 10, 12, and 14 d after tumor inoculation. For DMXAA administration, mice were injected i.t. with 20 µg DMXAA either 9 or 13 d after tumor inoculation.

Depletion antibodies (anti‐CD4, anti‐CD8, and anti‐NK1.1) as well as anti‐PD‐1 (clone RMP1‐14‐CP157), anti‐CTLA4 (clone 9D9), and anti‐CD47 (clone MIAP301) were purchased from BioXCell. The MitoSOX, CCF4‐AM, and MitoTEMPO reagents were purchased from Thermo Fisher Scientific and used according to the manufacturer's instructions. Cytochrome C (Cyt‐C), N‐acetylcysteine (NAC), and β‐lactamase were purchased from Millipore‐Sigma. The Annexin‐V staining kit was purchased from Cedarlane laboratories. The Accum™ and its leucine mutant derivative were synthesized as previously described.

Tumor Immunotherapy in Murine Models

Check if the same lab product or an alternative is used in the 5 most similar protocols
For studies with MC38 tumors, wild type C57BL/6 mice were implanted with MC38 cells as described above. When tumor volume reached an average of 75 mm3, mice were randomized into groups of n = 15 mice. For studies with E0771 tumors, Ptpn6fl/fl mice were implanted as described above. For anti-PD1 studies: mice were dosed intraperitoneally (IP) with anti-PD1 (clone RMP1-14, BioXCell) or rat IgG2a isotype control (clone 2A3, BioXCell) at 10 mg/kg twice weekly (BIW). For anti-CD47 studies, mice were dosed IP with anti-CD47 (clone miap301, BioXCell) or rat IgG2a isotype control (clone 2A3, BioXCell) at 20 mg/kg BIW for 2 weeks as described in Liu et al. (41 (link)).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
