NCI-h716 cells were seeded in RPMI 1640 (Hyclone, Logan, UT, USA) containing 10% fetal bovine serum (Hyclone, Logan, UT, USA) and used for all experiments. Cells were incubated in humidified air with 5% CO2 at 37 °C. Cells were transfected with Opti-MEM medium (Invitrogen CA, USA) and lipo2000 (Invitrogen CA, USA). Cells were divided into three groups: 25 mM High glucose (HG) group, HG + Emodin (100 μM) (Cat No. JOT-10023, Chengdu Pufei De Biotech Co., Ltd) group, and HG + Emodin + siRNA/inhibitor group. For the experiments involving silence of TCF7L2, cells were transfected with TCF7L2-targeting siRNA oligonucleotide synthesized by Shanghai GenePharma Co., Ltd. For the experiments involving SB216763 (a specific GSK3β inhibitor; ab120202; Abcam, Cambridge, MA, USA), cells were incubated with the agent at 37 °C for 24 h.

Aloe emodin (1 ), catechin (2 ) chrysophanol (3 ), emodin (4 ), emodin-8-O-β-d -glucoside (5 ), physcion (6 ), rhein (7 ), sennoside A (8 ) and sennoside B (9 ) were purchased from Chengdu Pufei De Biotech Ltd. (Chengdu, China); the purity of all reference compounds was > 98%. Emodin and Captopril Ameliorate TAC-Induced Cardiac Damage

Mice were fed for one-week acclimatization. They were then randomly divided into control, transaortic constriction (TAC), TAC+emodin (10, 20, and 40 mg/kg/day, 0.1 mL/10 g) and TAC+captopril (10 mg/kg/day, 0.1 mL/10 g) groups (10 mice in each group). Captopril (Xudong Haipu Pharmaceutical Co., Ltd., Shanghai, China) and emodin (≥98% in purity, Chengdu Pufei De Biotech Co., Ld., Chengdu, China) were dissolved in distilled water before use. The dosages of emodin and captopril were determined according to previous studies19 , 20 (link), 21 (link). Animals were anaesthetized with sodium pentobarbital (6 mg/kg, i.p., Sigma, St. Louis, MO, USA) and the effective anesthesia was verified by a loss of interdigital reflex. Next, TAC was performed as previously described18 (link). emodin or captopril was intragastrically administered into animals daily for four consecutive weeks. Control animals were given an equal volume of saline.

(Chengdu, China). Torachrysone [(TCS), ≥ 98%] was purchased from Yunnan Xili biology science and technology co., ltd (Kunming, China). The dried root of PM was collected from the Luoshiwan Chinese Medicine Market (Kunming, China). Dried PM decoction pieces were verified by Professor Jie Yu. PM credential specimens (No. 19080322) were stored in the Key Laboratory of Southern Medicine Utilization, Yunnan University of Chinese Medicine (Kunming, China). The other chemicals and reagents used were described in our previous report[10 (link)].
