Bacterial DNA Extraction and Electrophoresis

Total DNA was extracted from the bacterial DNA extraction as described by Seldin and Dubnau [42 (link)] using phenol-chloroform. After extraction, the DNA sample was mixed with blue juice dye in a proportion of 1:2 of the total volume of the mixture and subjected to horizontal electrophoresis in 1 % (w/v) agarose gel. Ethidium bromide (0.1%) was added to 10× diluted Tris-acetate-EDTA (TAE) buffer for 30 min at a constant voltage of 90 V. The molecular weight marker used was the “1 Kb Plus DNA Ladder” (Promega®, Madison, USA). The gel was observed and photographed under UV light using a UV light transilluminator (model) to analyze the purity and quality of the genetic material.

Anaplasma 16S rRNA Gene PCR
