Immunoprecipitation of Cellular Proteins

Cells were harvested and lyzed using NP-40-based lysis buffer with protease and phosphatase inhibitors. Total protein (500 μg) in 1 ml lysate was incubated with 2 μg of anti-NHE1 antibody (Abcam, UK) or anti-FLT3 antibody (Santa Cruz Biotechnology, TX, USA) [45 (link)] at 4 °C overnight. The antibody-protein complex was pulled down using Protein G sepharose (ThermoFisher Scientific) at 4 °C for 2 hours. The sepharose was washed twice with lysis buffer and eluted by SDS protein loading dye by boiling for 5 min. The supernatant was collected and stored at −80 °C for further analysis.
