Chondrocyte Pellet Implantation in Nude Mice

All experiments were performed according to the protocol approved by the Animal Care and Use Committee of the University of Tokyo. Immunocompromised BALB/cAJc1-nu/nu mice (6 weeks old, male) were obtained from Nisseizai, Tokyo, Japan. Under general anesthesia application with Isofluorane (Sigma–Aldrich), a 10 mm left dorsal skin incision was made in the nude mouse. The chondrocyte pellets cultured in vitro for 3 weeks were harvested and inserted subcutaneously. Four weeks after the implantation, the mice were euthanized, after which the pellets were harvested. Blood vessels and excess tissues were trimmed from the pellets, and photographed before being subjected to histological and biochemical analysis.

Cannabinoid and Opioid Receptor Agonist Protocol

JWH015, a cannabinoid 2 receptor (CB2) agonist (CB2 Ki = 13.8 nM, 30 to 80 fold selectivity versus CB1) was obtained from Cayman Chemical (Ann Arbor, MI). JWH015 was dissolved in a vehicle solution of 10% dimethyl sulfoxide, 10% Tween-80, and 80% saline (Sigma, St. Louis, MO). The mu-opioid agonist (MOR) morphine sulfate was purchased from the NIDA Drug Supply program (Rockville, MD) and was dissolved in saline. Formalin solution was obtained by diluting 1.5% of formaldehyde in saline. Ketamine:xylazine (80 mg/kg: 12 mg/kg; Phoenix Pharmaceutical, St. Joseph, MO) was used to anesthetize animals in order to insert microdialysis probes into the nucleus accumbens. The antibiotic gentamicin (Phoenix Pharmaceutical, St. Joseph, MO) was provided as a single subcutaneous dose (8mg/kg). Drugs were weighed out and dissolved in vehicle daily, prior to use. Cocaine hydrochloride was purchased from the NIDA Drug Supply program (Rockville, MD) and used as a positive control in microdialysis and HPLC studies. Finally, 5% and 2.5% isofluorane (Sigma, St. Louis, MO) mixed in 2.5 L/min of oxygen was delivered through a nose cone and used to induce and maintain anesthesia, respectively, for paw incision and SNI surgeries.

Establishment and Monitoring of Murine Tumor Models

For experiments with EL4-derivatives, mice were challenged with 1 × 106 cells s.c. in the flank. For B16.F10, 1 × 105 cells were used, for CT26, 2 × 105 cells, and for TC-1, 1 × 105 cells. Engraftment was on one or both flanks, as indicated in the text. When average tumour size reached 36 mm2 (calculated as the product of the two bisecting diameters), mice were subjected to intratumoural therapy; mice were anesthetized with isofluorane (Sigma-Aldrich) using a SomnoSuite anesthesia system (Kent Scientific), and compounds injected using 29 G needles. For some experiments, compounds were injected s.c. peritumourally, or s.c. on the contralateral limb from the tumour site. tumour growth was monitored every 1–2 d until ethical endpoint, when tumour size on either flank exceeded 200 mm2. Survival was recorded as time from tumour implantation to ethical endpoint.

Characterization of Pharmaceutical Compounds

Acetonitrile and methanol were high-performance liquid chromatography (HPLC) grade. Clear glass HPLC vials with crimpable lids and 0.45-µm nylon filter papers, SlowFade Gold Antifade Mountant with DAPI, and optimum cutting temperature (O.C.T.) solution were all purchased from Fischer Scientific. Tetracaine base and aciclovir base were both BP grade (99.9%). Isofluorane, isopentane, carrageenan, sterile saline, hematoxylin (Gill No3) solution, eosin solution, and differentiating solution were supplied by Sigma-Aldrich. Diclofenac diethylamine BP grade (99.9%) was from Unique Chemicals. Certified dextran standards (12 kDa, 80 kDa, and 150 kDa) were supplied by Merck Life Science. Concentrated hydrochloric acid and sodium hydroxide were from Fluka. Sodium acetate was provided by Alfa Aesar. Deionized water was obtained by purification using an Elgstat water purifier (Elga Ltd). Hydroxypropyl methylcellulose grade 65SH viscosity 50 cP with the brand name Metolose was supplied by Shin-Etsu Chemical Ltd.
