Tunicamycin (T7765), Phorbol 12-myristate 13-acetate (PMA) (P8139), LPS (L2630), and ATP (A6419) were purchased from Sigma-Aldrich while nigericin (tlrl-nig) was obtained from Invivogen. IRE1 inhibitor MKC8866 was provided by Fosun Orinove.

Recombinant TGF-β3 for Cell Stimulation

Recombinant human TGF‐β (TGF‐β3; #243‐B3‐010) was obtained from R&D Systems (Minneapolis, MN, USA). Nigericin (#tlrl‐nig) was purchased from InvivoGen (San Diego, CA, USA), and lipopolysaccharide (LPS) (#L4391) was from Sigma‐Aldrich (Merck, Darmstadt, Germany). Z‐VAD‐FMK (#G7232) was from Promega (Madison, WI, USA), and Z‐DEVD‐FMK (#FMK004) was from R&D Systems. SB202190 (#ab120638) was from Abcam (Cambridge, UK), and SB203580 (#tlrl‐sb20) was from InvivoGen. LY364947 was from Calbiochem (#616451; Merck).

Characterization of Pyroptosis Signaling

The primary antibodies used were against IL-1β (AF-401-NA, R&D), caspase-1 (AG-20B-0042-C100, Adipogene), GSDMD (ab210070, Abcam), MLKL (generated in house (74 (link)), available from Millipore, MABC604, clone 3H1) and β-actin (A-1978, Sigma-Aldrich). Flow cytometric staining utilized the following antibodies from BioLegend; purified anti-mouse CD16/32 antibody (101302), APC-conjugated anti-Ly6G (127614), FITC-conjugated anti-F4/80 (123108), PE-conjugated anti-B220 (103208), APC-CY7-conjugated anti-CD3 (100222) and Brilliant Violet 421-conjugated anti-CD11b (101236). 123 count eBeads were purchased from Thermo scientific (01-1234-42). Ultrapure LPS (tlrl-smlps) and nigericin (tlrl-nig) were purchased from InvivoGen. MSU was generated in house, as previously described(63 (link), 75 (link)). Imject™ Alum Adjuvant (77161, thermo scientific), Z-VAD-fmk (187389-52-2, Santa Cruz Biotechnology), CA-074-Me (sc-214647, Santa Cruz Biotechnology), necrosulfonamide (ab143839, Abcam), potassium chloride (P954, Sigma-Aldrich) and glycine (G7126; Sigma-Aldrich) were purchased as indicated.
