Pmx ires blasticidin vector

Manufactured by Cell Biolabs

The PMX-IRES-Blasticidin vector is a bicistronic mammalian expression vector that allows for the co-expression of a gene of interest and the blasticidin resistance gene. The vector contains an internal ribosome entry site (IRES) sequence that enables the translation of the blasticidin resistance gene from the same mRNA as the gene of interest, facilitating the selection of successfully transfected cells.

Stable miR-10b Expression in RDEB-KCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
For stable expression of miR-10b in E6/E7 immortalized RDEB-KCs, the human MIR10B gene was cloned into the pMX-IRES-Blasticidin vector (Cell Biolabs Inc., RTV-016), downstream of the constitutive Pol-III U6 promoter. Primer sequences are given in Supplementary Table S3 in Additional File 1. All constructs were analyzed using Sanger sequencing before viral packaging.
Viral particle production using pMX_U6_miR10b was done as described previously [26 (link)]. Expression and maturation of miR-10b was confirmed by TaqMan qPCR (Supplementary Fig. S1C-E in Additional File 1).

Stable Expression of miR-10b in Keratinocytes

Check if the same lab product or an alternative is used in the 5 most similar protocols
For stable expression of miR-10b in E6/E7 immortalized HC-KCs and RDEB-KCs, the human MIR10B gene was cloned into the pMX-IRES-Blasticidin vector (Cell Biolabs Inc., RTV-016), downstream of the constitutive Pol-III U6 promoter. Primer sequences are given in Supplementary Table S3. All constructs were analyzed using Sanger sequencing before viral packaging. Viral particle production using pMX_U6_miR10b was done as described previously (26) . Expression and maturation of miR-10b was confirmed by TaqMan-PCR (Supplementary Fig. S1C-E).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
