Mtb Infection Dynamics in Ifnar1-/- Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mice, 129S2 (129SvPas), Ifnar1−/− in 129S2 background 24 (link), and Ifnar1−/−44 (link) in C57BL/6 background were bred and kept under specific pathogen-free conditions at the Max Planck Institute for Infection Biology in Berlin, Germany. C57BL/6 animals were purchased from Charles Rivers Laboratories. Mice were 10–12 weeks of age at the beginning of the experiments. Aerosol infection with Mtb strain H37Rv and enumeration of bacteria in lung tissue were performed as previously described 34 (link). To generate GFP-expressing Mtb, competent H37Rv was transformed with the plasmid pGFM-11 by electroporation and selected on kanamycin 45 (link). Chimeric mice were obtained by reconstituting lethally irradiated mice with BM cells. Mtb infection was performed 8–10 weeks following transplantation. PMNs were depleted by administration of 250 μg mAb Ly6G (clone 1A8, BioXCell) i.p. for 1 week at 2-day intervals, starting 7 days p.i. All experiments were performed in accordance with German Animal Protection Law, in Berlin, by the State Office of Health for Social Affairs in Berlin (Landesamt für Gesundheit and Soziales, LAGeSo; approvals G0310/06, G0221/11, T0223/11, and T0087/13).

Lung Tissue Immunohistochemistry for Influenza

Check if the same lab product or an alternative is used in the 5 most similar protocols
Lung tissues from two animals per treatment group and time point were processed and stained as previously described (9 (link)). Antibodies specific for PR8 hemagglutinin protein (catalog number NR-3148; BEI Resources) and Ly6G (clone 1A8; Bio X Cell) were detected with immPACT vector red and diaminobenzidine immPACT DAB, respectively (Vector Laboratories).

Functional grade purified antibodies against CD4 (16-0042, clone RM4-5 and clone GK1.5, Rat IgG2bK; eBioscience), TSLP (MAB555, clone 152614; R&D System, 16-5491, clone eBio28F12; eBioscience ), NKG2D (catalogue number BE0111; BioXCell), LY6G (clone 1A8; BioXCell), IL-4 (catalogue number BE0045; BioXCell), and appropriate isotype controls (BioXCell Rat IgG2a, catalogue number BE0089 for TSLP and Ly6G; Rat IgG2b, catalogue number BE0090 for CD4; hamster IgG catalogue number BE0091 for NKG2D; Rat IgG1, catalogue number BE0088 for IL-4) were injected intraperitoneally at a dose of 0.1 mg/mouse. Dexamethasone 21-phosphate disodium salt (DXM) (D1159; Sigma) was injected at 10 mg/kg mouse body weight, the same volume of PBS being injected in control mice.

Ly6G Depletion in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mice were injected with mAb depleting Ly6G (clone 1A8) that was purchased from Bio-X-Cell (cat# BE0075-1) and administered IP in doses of 500μg one day prior to eCPMV treatment and then once weekly for the duration of survival experiments. Greater than 95% depletion of target cell populations in the lung was confirmed by flow cytometry (Supplementary Fig. 9).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
