Flow Cytometric Analysis of EMT in NCCs

EMT in NCCs was analyzed by flow cytometry. NCCs were harvested, fixed with 4% paraformaldehyde, and permeabilized with 0.1% Triton X-100 in phosphate-buffered saline (PBS) for 20 min at room temperature. After that, the cells were blocked by 5% goat serum in PBS for 30 min and then incubated with E-cadherin Rabbit mAb (PE conjugate) (Cell Signaling, Beverly, MA, USA), Vimentin Rabbit mAb (Cell Signaling, Beverly, MA, USA) for 30 min at 37°C, and then incubated with fluorescein isothiocyanate (FITC)-labeled goat anti-rabbit IgG secondary antibody (Abcam, Cambridge, MA, USA) for 30 min at 37°C. Flow cytometry analysis was performed on a BD FACSCalibur system. The average fluorescence intensity was analyzed using FlowJo 7.6.1 software.
