Immunoblotting Analysis of Cellular Proteins

Cells or tissues were collected/homogenized and lysed on ice in an RIPA buffer (25 mM Tris–HCl, pH 7.5, 150 mM NaCl, 1 mM EDTA, 1% NP-40 and 5% glycerol) with protease inhibitors (#88660SPCL, Thermo Fisher Scientific) and a phosphatase inhibitor mix (sc-45044, Santa Cruz). Western Blot Analysis of Exosomal Proteins

Total protein of indicated cells or isolated exosomes was extracted using RIPA Lysis Buffer (Beijing, China) at 4°C for 30 min. Protein concentration was determined by PierceTM BCA Protein Assay Kit. Proteins were then concentrated on SDS-PAGE (6%) and separated by SDS-PAGE (12%), and transferred to nitrocellulose filter membranes. After blocking with 3% BSA, membranes were subsequently incubated with primary antibody anti-HuR (sc5261, Santa Cruz), anti-GM130 (11308-1-AP, Proteintech), anti-TSG101 (ab83, Abcam), anti-CD9 (ab92726, Abcam), anti-Lamp2b (ab18529, Abcam) and anti-GAPDH (D110016-0100, BBI Life Sciences). After washing three times in TBST, the membranes were incubated with horseradish peroxidase-conjugated secondary antibody at room temperature for 1 h.

Apoptotic Signaling Pathways in Cell Culture

All cell culture media, supplements and modifying enzymes were purchased from Invitrogen (Karlsruhe, Germany) if not otherwise indicated. Doxorubicin was from Biotrend Chemicals (Cologne, Germany), paclitaxel, and propidium iodide were obtained from Sigma-Aldrich (Taufkirchen, Germany). Gö6983, KU55933, SB203580, U0126 and Chk2 inhibitor II were from Merck (Darmstadt, Germany). The caspase inhibitors Z-VDVAD-FMK and Z-VAD-FMK were derived from R & D Systems (Wiesbaden, Germany). The following antibodies were used in this study: anti-HuR (sc-5261) and anti-Lamin B1 (sc-20682) from Santa Cruz (Heidelberg, Germany), anti-caspase-2 (#611022, BD Biosciences, Heidelberg, Germany), anti-caspase-3 (#9662), anti-caspase-7 (#9494) anti-PARP (#9542) and anti-Bid (#2002) antibodies were from Cell Signaling (Frankfurt, Germany), and anti-β-actin (#A2228) from Sigma-Aldrich. The goat anti-rabbit (sc-2054) and goat anti-mouse (sc-20559) HRP-linked antibodies were from Santa Cruz and Alexa Fluor 488 goat anti-mouse was from Life Technologies (Darmstadt, Germany). Radionucleotides were from Perkin Elmer (Rodgau, Germany) and the ECL system and Hyperfilms were from GE Healthcare (Freiburg, Germany).
