Quantification of Midazolam and Metabolite

Plasma concentrations of midazolam and 1′-hydroxymidazolam were quantified using HPLC-MS/MS on a system consisting of a Surveyor HPLC autosampler, Surveyor MS quaternary pump, and TSQ Quantum Discovery triple quadrupole mass spectrometer (Thermo Scientific, San Jose, CA). After liquid-liquid extraction, midazolam and metabolite concentrations were measured simultaneously with electrospray ionization in the positive mode as described previously.13 (link) midazolam, midazolam d4, 1′-hydroxymidazolam, and 1′-hydroxymidazolam-d4 were purchased from Cerilliant (Round Rock, TX). The lower limit of quantification for both midazolam and 1′-hydroxymidazolam was0.2 ng/ml, and the accuracy and precision were within 7%.

N-Hexane and ethyl acetate were purchased from Merck (Milan, Italy). β-glucuronidase from Escherichia Coli was purchased from Merck (Milan, Italy).

Characterization of Metabolic Enzyme Modulators

The pMIR-REPORT plasmid was from Life Technologies (Grand Island, NY) and the Renilla luciferase vector pRL-CMV was purchased from Promega (Madison, WI). All synthesized DNA oligos used in this study were obtained from Integrated DNA Technologies, Inc. (Coralville, IA). Lipofectamine RNAiMAX was from Life Technologies and the transfection reagent DharmaFECT Duo was purchased from GE Dharmacon.

Tacrolimus and Midazolam Metabolism

Midazolam Pharmacokinetic Analysis

Six different lots of drug-free (blank) human plasma prepared with lithium heparin as an anti-coagulant were obtained from BioreclamationIVT (Baltimore MD). HPLC grade methanol and acetonitrile were purchased from Fisher-Scientific (Pittsburgh, PA, USA). Deionized water from Barnstead Nanopure™ water purifying system (Thermo Fisher Scientific, Marietta, OH, USA) was used for all experiments.

Metabolic Drug Interaction Profiling

(Manchester, England) and nicotinamide adenine dinucleotide phosphate (NADPH) from AppliChem (Darmstadt, Germany). HLM from 50 individual mixed-sex donors were purchased from Xenotech (Lenexa, KS). Human hepatocytes were isolated in house from liver tissue obtained from a female donor undergoing surgical resection at the Department of Surgery, Uppsala University Hospital (Sweden) and cryopreserved as described previously34 (link),35 (link). Ethical approval was granted by the Uppsala Regional Ethics Committee (ethical Approvals Nos 2009/028 and 2011/037). Donors gave informed consent and all studies were performed in accordance with the current national regulations and ethical guidelines.
