ATP-Induced Current Measurement Protocol

ATP, ADP, UDP, GDPβS and bupivacaine hydrochloride were purchased from Sigma. Pertussis toxin (PTX), forskolin, tetraethylammonium (TEA), quinine, and 4-aminopyridine (4-AP) were purchased from Tocris. Stock solutions of all drugs were made in water and diluted to the appropriate working concentrations in ACSF. Drugs were applied to the slices either through bath application or using a picopump (WPI pneumatic picopump, Sarasota, FL). The diameter of the drug application pipette tip was ∼3–4 μm. The pressure (10 psi) and duration (100 ms) of the puff was controlled and the distance between the patched cell and puff pipette was kept constant (∼15 μm). This was achieved by marking the position of the two pipettes (recording and puff) on the display screen and adjusting the distance of the puff pipette until the preferred distance was reached. The holding pressure of the puff pipette was maintained at -2 psi to prevent leakage, but there may still be minimal spontaneous leakage. For experiments involving testing of antagonists on ATP-induced current, control applications of ATP were performed and then on the same cell the effect of the antagonist was tested.

Pharmacological Intervention in Pain Study

Tetraethylammonium, glibenclamide, apamin, and charybdotoxin were purchased from Tocris (Bristol, UK). DMSO and Tween 20 were purchased from Sigma-Aldrich (St. Louis, MO, USA). Naloxone hydrochloride, β-funaltrexamine, naltrindole, nor-binaltorphimine, DMSO, and Tween 20 were bought from Sigma-Aldrich (St. Louis, MO, USA). Morphine sulphate was purchased from Lipomed (Cambridge, MA, USA). All drugs were dissolved in 0.9% NaCl. The vehicle consisted of DMSO, Tween 20, and 0.9% NaCl at a ratio of 5:5:90. All treatments were administered either intraperitoneally or subcutaneously at a volume of 10 mL/kg of body weight. Intraperitoneal injection was administered in the intraperitoneal cavity and subcutaneous administrations were made into the loose skin over the interscapular area.

Pharmacological characterization of vascular responses

Equilin, 17β-estradiol, phenylephrine, acetylcholine, indomethacin, L-NAME (Nω-nitro-L-arginine methyl ester), and KCl were purchased from Sigma-Aldrich (St. Louis, MO, USA). Tetraethylammonium, glibenclamide, 4-amynopiridine, ouabain, (S)-(−)-Bay K 8644, U46619 (9,11-dideoxy-11α,9α-epoxymethanoprostaglandin F2α), ICI 182,780, KT 5720, KT 5823, SQ 22,536 (9-(tetrahydro-2-furanyl)-9H-purin-6-amine), and ODQ (1H-[1 (link)–2 (link),4 (link)]oxadiazolo [4,3-alpha]-quinoxalin-1-one) were obtained from Tocris Bioscience (Bristol, UK). Caffeine was purchased from Roth (Karlsruhe, Germany).
Equilin and 17β-estradiol were dissolved in 70% v/v ethanol with further dilution in distilled water before use. Working concentrations of ethanol in the bath were < 0.01% (v/v).
