The endovascular perforation was produced as previously described with modifications8 (link). Anesthesia was induced with sodium pentobarbital (80 mg/kg). After anesthesia of intraperitoneal injection in mice, a laser Doppler flowmetry probe (Moor Instruments Inc., Wilmington, DE) was adhered at the left temporal bone to monitor the blood flow of the left hemispherea. The left common carotid artery (CCA), internal carotid artery (ICA) and external carotid artery (ECA) were exposed and the external carotid artery (ECA) was fashioned into a 3-mm stump. A 6-0 monofilament nylon suture was advanced into the internal carotid artery (ICA) from the ECA stump until resistance was felt, about 1.0 cm from Common carotid artery bifurcation have resistance to insert again after 2 mm pricking blood vessels and then the suture was immediately withdrawn to induce subarachnoid hemorrhage. Cerebral Blood Flow Measurement

After evaluating the neurological deficit in the second rat experiment, CBF during cerebral ischemia was measured with a laser Doppler flowmetry probe (Moor Instruments Ltd). In brief, under anesthesia, a small incision was made in the center of the skull. Remove the underlying fascia and press it until there was no bleeding point on the skull. A small area of the skull was exposed to allow placement of the laser-Doppler probe.

Transient Focal Cerebral Ischemia in Rodents

Middle cerebral artery occlusion. Temporary focal cerebral ischemia was induced under general anesthesia (isoflurane 1.5-2%) by occlusion of the right middle cerebral artery (MCAO) using the intraluminal filament technique. Briefly, a nylon thread (80 μm in diameter) with a distal cylinder (1.5 mm and 180 μm in diameter) was inserted into the common carotid artery, advanced to the origin of the MCA (exept for sham) and removed 60 min later to allow reperfusion. A laser-Doppler flowmetry probe (Moor Instruments) was used to continuously monitor CBF. Post-surgery analgesic care comprised intradermal ropivacaine (25 μL at 2.5 mg/mL, Naropeine) around sutures and applications of lidocaine/prilocaine cream (2,5% EMLA, AstraZeneca). The animals were then allowed to recover and were killed at day 2 or 28 after MCAO.
