Cell Viability Assay with MTT

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells were plated on 96-well plates (8000 per well) with a volume of 100 µL cell culture medium on day 0. MABS treatment was performed on day 1. Then, 3 h before the indicated time points (4–72 h) the MTT-reagent was applied and incubated. Readout was performed on a Tecan Sunrise Absorbance microplate reader. Data analysis was performed with GraphPad Prism software (La Jolla, CA, USA).

Quantifying Intracellular Glutathione

Check if the same lab product or an alternative is used in the 5 most similar protocols
Intracellular GSH was determined with a quantification kit (Dojindo Laboratories) according to the manufacturer’s instructions. Briefly, MEFs were lysed by the addition of 80 μL of 10 mM HCl and three freeze-thaw cycles. To the homogenate, 20 μL of 5% salicylsalicylic acid (SSA) was added, and the mixture was centrifuged at 8000× g for 10 min. GSH levels in the supernatant were determined according to the manufacturer’s protocol by measuring absorbance at 405 nm with a Sunrise absorbance microtiter plate reader (Tecan Japan, Kanagawa, Japan).

Growth Characterization in Minimal Media

Check if the same lab product or an alternative is used in the 5 most similar protocols
Phenotypes were reconfirmed by testing their growth in liquid media in a 96-well plate. Overnight incubations of the strain of interest were subcultured in LB and grown until an OD600 of 0.4 was reached. Cells were subsequently washed in MOPS minimal media with no carbon source and diluted 1:100 into the carbon source of interest. OD600 measurements were taken every 15 min for up to 48 h using a Sunrise absorbance microplate reader (Tecan Life Sciences).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
