Characterization of Immune Cell Populations

Type II collagen (CII) was purchased from Chondrex (Redmond, WA, USA). Hyaluronidase was obtained from Sigma-Aldrich (St. Louis, MO, USA). Phorbol 12-myristate 13-acetate (PMA) and ionomycin were all products from eBioscience (San Diego, CA, USA). Fluorochrome-conjugated anti-human monoclonal antibodies (mAbs) including CD4 (clone: RPA-T4), CD14 (clone:M5E2), CD19 (clone: HIB19), ICOS (clone:C398.4A), ICOSL (clone:2D3) were from Biolegend (San Diego, CA, USA). Fluorochrome-conjugated anti-mouse mAbs including CD3 (clone:145-2C11), CD19 (clone:6D5), NK1.1 (clone:PK136), ICOS (clone:15F9), ICOSL (clone:HK5.3), CD44 (clone:IM7), B220 (clone:RA3-6B2), CD138 (clone:281-2), GL7 (clone:GL7), CD25 (clone:3C7), Foxp3 (clone:150D), CXCR5 (clone:L138D7), IFN-γ (clone:XMG1.2), TNF-α (clone:MP6-XT22), IL-4 (clone:11B11), IL-9 (clone:RM94A), IL-17 (clone:TC11-18H10.1) and IL-22 (clone:poly5164) were from Biolegend. Fluorochrome-conjugated anti-mouse mAbs CD4 (clone: GK1.5), CD38 (clone:90), CD95 (clone:15A7), CD62L (clone: MEL-14), IL-6 (clone:MP5-20F3), IL-21 (clone: FFA21) were from eBioscience. Anti-mouse CD11b mAb was from Mitenyi Biotec (Bergisch Gladbach, Germany). The anti-mouse ICOSL blocking antibody (clone HK5.3) was from Biolegend.

Monoclonal Antibody Staining for Flow Cytometry

The following monoclonal antibodies conjugated with fluorochrome were used: Desmoglein-2(Santa Cruz Biotechnology), 41BBL, ICOSL (BioLegend), OX40L, CAR, CD46 (BD Biosciences), FAP (R&D Systems). Cells were stained with these antibodies for 30 min at 4 °C. Live/dead discrimination was determined via inclusion of 7-aminoactinomycin D (7AAD; BD Pharmingen). Stained cells were analyzed using Gallios flow cytometer with Kaluza software (BD Biosciences) according to the manufacturer’s instructions.

Antibody Reagents for B Cell Immunology

Antibodies against mouse CD4, CD19, CD95, GL-7, ICOS, ICOSL, PD-1, and B220-647 were purchased from Biolegend, CXCR5, B220, IgG1, IgG2a, IgG2b, and IgG3 antibodies from BD Biosciences, Bcl-6, XBP-1, and IRF-4 from Santa Cruz, and BAFF (1C9) from Enzo. Anti-μ F(ab)2 was purchased from Jackson ImmunoResearch. Anti-μ (clone B7.6), anti-NP (clones H33L and B1-8), 2.4G2 (FcγRIIb /FcγRIII block) and Ac38 idiotype antibodies were purified from hybridoma supernatants. (Ac38 is an idiotype antibody that recognizes B-1-8 specificities generated during NP immunization). Recombinant mouse BR3-Fc and isotype control protein were generated using mammalian expression systems and standard purification protocols. H33L and B1-8 were gifts from Dr. Garnett Kelsoe (Duke University). IL-4 and IL-5 were purchased from Peprotech, recombinant BAFF (rBAFF) from R&D Systems, NP-OSu from Biosearch Technologies, KLH and PNA-biotin from Sigma Aldrich, alum from Thermo Scientific, and streptavidin-Alexa 488 and Alexa 647 from Invitrogen. Streptavidin-alkaline phosphatase and anti-IgG alkaline phosphatase were purchased from Southern Biotech. The Fc-binding TG19320 peptide was synthesized as described (39 (link), 40 (link)).

Costimulatory Marker Profiling in Murine B Cells

IDO2 wt and ko C57BL/6 spleen cells were harvested and stimulated for 72h with anti-IgM F(ab′)2 (Jackson Immunoresearch, 20μg/ml) or anti-CD40 (eBioscience, 2μg/ml) + IL-21 (eBioscience, 100ng/ml) in IMDM + 10% FCS + 5μM 2-mercaptoethanol, 2mM glutamax (Gibco), and 50μg/ml gentamycin. Levels of the following costimulatory markers were directly measured by flow cytometry on a BD FACSCanto with subsequent analysis using FlowJo Software (TreeStar): CD40 (eBioscience), CD80 (eBioscience), CD86 (eBioscience), ICOSL (BioLegend), IL-21R (BD Biosciences), OX40L (BioLegend), PDL1 (BioLegend), and MHC Class II (eBioscience). A minimum of 3 replicate mice were analyzed per condition.
