U46619, N-(methylsulfonyl)-2-(2-propynyloxy)-benzenehexanamide (MSPPOH), medotomidine hydrochloride, and atipamezole hydrochloride were ordered from Cayman Chemicals. BAPTA-tetrapotassium and Alexa 488 hydrazide were obtained from Invitrogen, isoflurane from Fresenius Kabi Canada Ltd., Pluronic F127 and Rhod-2/AM from Biotium, buprenorphine from Champion Alstoe Animal Health, and enrofloxacin from Bayer, meloxicam from Boehringer Ingelheim, ketamine from Vétoquinol N.-A. Inc. AAV2/5.GfaABC1D.GCamP6f and AAV9.hSyn.GCaMP6f were ordered from Penn Vector Core, PA, USA, pZac2.1-GfaABC1D-mCherry-hPMCA2w/b plasmid was ordered from Addgene.

Noradrenaline Pharmacological Manipulation

Noradrenaline bitartrate was dissolved in distilled water at a stock concentration of 100 mM and diluted in ACSF to 10 μM. Prazosin hydrochloride (Cayman Chemical, Ann Arbor, MI #15023) was dissolved in pure dimethyl sulfoxide at a 10 mM stock concentration and diluted to a final volume of 10 μM in ACSF. Atipamezole hydrochloride (Cayman Chemical, #9001181) was dissolved in distilled water at a stock concentration of 50 mM, sonicated for 2 minutes, and diluted to a final concentration of 10 μM in ACSF. Recordings were performed in the continuous presence of picrotoxin (50 μM). All solutions equilibrated at 34 ± 0.5 °C prior to slice application.
