Intracellular cAMP and PKA Assay in A. flavus

To assay the intracellular cAMP levels in the A. flavus strains, the liquid shaken mycelial were harvested after 48 h of inoculation and lyophilized overnight. The free dry mycelium was used for total protein extraction following the protocol as previously described [24 (link),39 (link)]. To measure the PKA activities, the PepTag® Assay for Non-Radioactive Detection of the cAMP-Dependent Protein Kinase kit (Promega, Madison, WI, USA) was utilized.
