Puromycin, 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), DCA, linolenic acid (LA), spermine, cisplatin (CDDP), 5-fluouracil (5-FU), Paclitaxel (TAXOL), curcumin, epigallocatechin gallate (EGCG), silibinin (INN), metformin, ERK1/2 inhibitor PD98059 and glutaminase inhibitor 6-diazo-5-oxo-L-norlucine (DON) were purchased from Sigma. PKB/Akt inhibitor MK-2206 was obtained from Selleckchem. HNSCC cell lines including SAS and HSC3 (tongue cancer), FaDu (hypopharyngeal cancer) and OECM-1 (Oral squamous cell carcinoma, OSCC) were obtained elsewhere [23] (link). Animal procedures were approved in accordance with the Institutional Animal Care and Use Committee (IACUC), National Yang-Ming University (NYMU) [23] (link), [25] (link). For xenografic tumor growth assay, cells were subcutaneously injected at the back of nude mice from National Laboratory Animal Centre and tumor size/mass was recorded. Clinical human HNSCC tissues (Supplementary Table 1) were obtained under approval of the Institutional Review Boards (IRB) of MacKay Memorial Hospital (IRB approval number: 12MMHIS178) and National Taiwan University Hospital (IRB approval number: 203106005RINC), Taiwan.

Xenograft Tumor Growth Assay of HNC

Check if the same lab product or an alternative is used in the 5 most similar protocols
Puromycin, 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), CDDP, 5-fluouracil (5-FU), and Extracellular signal-regulated kinase 1/2 (ERK1/2) inhibitor PD98059 were purchased from Sigma-Aldrich. Protein kinase B (PKB/Akt) inhibitor MK-2206 was obtained from Selleckchem. The source and culture conditions of HNC cells (all are Human PapillomaVirus negative (HPV)) from different origins as well as in HNC bearing xenografic tumour growth assay [under the approval of Institutional Animal Care and Use Committee (IACUC), National Yang Ming Chiao Tung University (NYCU) were described elsewhere [26 (link), 27 (link)]. In brief, the same kind of 106 control (shLuc) and TFAM silencing HNC cells were transplanted in opposite side of flank in the same male nude mouse (5–7 weeks old CAnN.Cg-Foxn1nu/CrlNarl from Academia Sinica N = 9–10). The tumour size was followed every week and at the 4th week post transplantation, tumours were excised and weighted. Clinical human HNC tumours and its adjacent normal tissues were obtained from the Department of Oral Maxillofacial Surgery, NYCU Hospital under approval of the Institutional Review Boards (IRB) of NYCU Hospital (IRB#: NYMUH 2018B003) and were preserved in RNAlater immediately after surgical resection for qRT-PCR analysis (Table S1).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
