Colon Cancer Spheroid Assay with IL33

The sphere assay was performed as described previously (22 (link)). Briefly, colon cancer cells were plated in ultralow attachment plates (Corning) in X-VIVO medium (Lonza) at a density of 1,000–5,000 viable cells/well. Colon cancer cells were treated with recombinant IL33 (PeproTech) or IL33-conditioned macrophages for different time points. The NFκB inhibitor, BAY11-7082; the P38 inhibitor, SB203580; the JNK inhibitor, SP600125; the MEK/ERK inhibitor, PD98059 (Cayman Chemicals), and the iNOS inhibitor, SMT (Aladdin), and anti-human ST2 antibody (R&D Systems) were added in sphere culture for 1 week. Modulating RCC Cell Apoptosis via IL-33
