Honey Authentication via Carbohydrate Profiling

Maltulose monohydrate (99.3%) was purchased from Chem-Impex Intl. (Wood Dale, IL). Sucrose (99.5%), α-lactose monohydrate (99%), maltose monohydrate (99%), palatinose hydrate (>98%), and D-(+)-turanose (>98%) were purchased from Sigma-Aldrich (St. Louis, MO). Phenylboronic acid (PBA, >97%) was obtained from Strem Chemicals (Newburyport, MA). Acetonitrile (HPLC grade) and ammonium hydroxide (35% solution in water) were purchased from Fisher Scientific (Pittsburgh, PA). High purity deionized water with 18.2 MΩ·cm resistivity was prepared using a Milli-Q filtration system (Merck Millipore, Burlington, MA). High-purity nitrogen was used as the sheath gas.
Honey samples were all US Grade A and consisted of Nature Nate’s (McKinney, TX) Natural 100% Raw Unfiltered Honey (honey #1), locally sourced raw honey from Union County, OH (honey #2), and Kirkland’s Signature (Costco Wholesale, Seattle, WA) Organic Raw Honey (honey #3).

Analytical Standards for HPLC Analysis

For the purpose of determinations, analytical purity reagents (analytical standards) designed for liquid chromatography were used: hydrochloric acid, formic acid, ethanol, and acetonitrile from Sigma Aldrich (Steinheim, Germany), and methanol from J.T. Baker Mallinckrodt Baker B.V. Holland. Buffers, ninhydrin, and a mixture of standards for amino acid identification were obtained from Sykam (Eresing, Germany), standardized for amino acid analyses in the physiological (native) range. Analytical standards of chlorogenic, caffeic, and ferulic acid for HPLC were obtained from Extrasynthese (Genay, France). Analytical standards D-(+)-glucose, D-(−)-fructose, D-(+)- sucrose, D-(+)- melezitose, D-(+)-turanose, D-(+)-trehalose, D-(+)-raffinose BioXtra, Ca, K, Mg, Fe, Cu, Mn, Zn, Al, Cd, and Pb were obtained from Sigma- Aldrich (Steinheim, Germany), and D-(+)-maltose standard was obtained from Toronto Research Chemicals. Carrez I solution, Carrez II solution (Sigma Aldrich, Steinheim, Germany), and deionised water from a deioniser from Hydrolab Polska HLP 5P were used.

Comprehensive Honey Composition Analysis

Most of the honey samples were collected from different regions of Poland (27 samples) from private beekeepers and manufacturers. Other samples were collected from Australia (3 samples), Romania (2 samples), Greece, Kameron, France, Czech Republic, Russia and Brazil (1 sample from each mentioned country). A full list of samples, including place of cultivation, year, manufacturer, and their physicochemical properties is presented in Table 1.
Standard purity ≥95% D-pinitol, myo-inositol, D-chiro-inositol, ononitol, bornesitol, allo-inositol, cis-inositol, epi-inositol, D-glucose, D-fructose, D-maltose, lactose, D-sorbitol, and D-(+)-turanose and trimethylsilylimidazole (TMSI) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Standards of sucrose, xylose, quebrachitol, neo-inositol, muco-inositol with purity ≥98%, and pyridine were bought from Avantor (Gliwice, Poland). DPPH (2,2-Diphenyl-1-picrylhydrazyl), trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid), and 70% EtOH, were purchased from Sigma Aldrich (St. Louis, MO, USA). Ultra-pure water was obtained from a Milli-Q water system (Millipore Bedford, MS, Boston, Massachusetts, USA).

Antioxidant and Cytotoxic Assays Protocol

Foetal bovine serum (FBS), L-gluta- mine, Hank's balanced salt solution (HBSS), trypsin-EDTA (ethylenediaminetetraacetic acid), a nonessential amino acid solution (2 mM), a penicillin/streptomycin solution (100 U mL -1 and 100 mg mL -1 , respectively) and DMEM (Dulbecco's modified Eagle medium) were from Hyclone (Logan, USA). All other solvents and reagents were of analytical grade and purchased from a common source. Water was treated in a Milli-Q water purification system (TGI Pure Water Systems, USA).
