Swine Mycoplasma Vaccine Development

Squalene oil was purchased from Alfa Aesar (Seoul, Korea), Span 80 from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan) and Cremophor ELP from BASF (Ludwigshafen, Germany). Two different grades of carbomers, C-971P NF and C-940, Montanide ISATM 201 and an antigen solution incorporating a PCV type 2 antigen and inactivated Mycoplasma hyopneumoniae (J101 strain, phosphatase (−), arginine hydrolysis (−), urease (−)) antigen were supplied from Komipharm Int’l Co., Ltd. (Siheung, Korea). DNA extraction kit and PCR premix were purchased from iNtron Biotechnology (Seongnam, Korea). Rabbit anti-Mouse IgG2a secondary antibody and horseradish peroxidase (HRP) was purchased from Thermofisher (Waltham, MA, USA), mouse IgG1 antibody (also known as goat anti-Mouse IgG1, HRP) from GeneTex (Hsinchu City, Taiwan) and goat anti-Mouse IgG (H+L) secondary antibody was purchased from Thermofisher (Waltham, MA, USA). The Mycoplasma hyopneumoniae antibody test kit was purchased from IDEXX (Westbrook, ME, USA). Other used reagents were of analytical grade.

Synthesis of DMAA-DMAEMA Copolymers

 Monomers, N,N-dimethylacrylamide (DMAA) (049-19185), N,N-dimethylaminoethyl methacrylate (DMAEMA) (044-16276), and the cross-linker N,N′-methylenebisacrylamide (M0506) were obtained from Wako Pure Chemical Industries Ltd. Sodium glucuronic acid monohydrate and chondroitin sulfate C sodium salt used for adsorption were purchased from Wako Pure Chemical Industries, Ltd. Other chemicals were of analytical grade or higher.

Droplet-Interface Bilayer Microfluidics

A zwitterionic lipid, PC (Avanti Polar Lipids, Alabaster, AL, USA), and a nonionic surfactant, sorbitan monooleate (Span80, Tokyo Chemical Industry Co., Tokyo, Japan), were dissolved in hexadecane (purity > 98%; Tokyo Chemical Industry Co., Tokyo, Japan) and used as the continuous oil phase. Milli-Q water (H20MB0124, Merck KGaA) was used as the aqueous phase. To visualize a lipid membrane covering a droplet, a fluorescent-labeled lipid, 1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine-N-(lissamine rhodamine B sulfonyl) (ammonium salt; Rho-PE, Avanti Polar Lipids) was used. Calcein (Sigma-AldrichSt. Louis, MO, USA), a water-soluble fluorescein, was used to analyze the function of α-hemolysin isolated from Staphylococcus aureus (Sigma-Aldrich) reconstituted in a DIB. The microfluidic device was developed using a photoresist (SU-8 3025, MicroChem Corp., Westborough, MA, USA) and a polydimethylsiloxane (PDMS) package (Sylgard 184, Dow Corning Toray Corp., Tokyo, Japan).

Luteolin-Enriched Topical Formulation

(St. Louis, MO, USA). 1,8-cineole (1,3,3-trimethyl-2-oxabicyclo [2.2.2]octane, CAS# 470-82-6) and nerolidol [(6E)-3,7,11-trimethyldodeca-1,6,10-trien-3-ol, CAS# 7212-44-4)] were purchased from Alfa Aesar (Haverhill, MA, USA). The MultiScreen® Permeability Filter Plate, 0.4 µm, non-sterile (The skin PAMPA kit) was from Merck (Darmstadt, Germany). All solvents used in this study were of analytical grade.

Quantitative Analysis of Bioactive Compounds

Fabrication of Polyvinylidene Fluoride Membrane

Non-woven polypropylene support (PP, Novatexx 2471) was purchased from Freudenberg-Filter (Weinheim, Germany). Polyvinylidene fluoride (PVDF, KYNAR HSV900) was provided by Arkema (Colombes, France) which was used as the membrane polymer. N, N-Dimethylacetamide (DMAc) (Purity ≥ 99.9%, Sigma-Aldrich, St. Louis, MO, USA) was utilized as the organic solvent for the preparation of the PVDF dope. The dopamine hydrochloride (DA, 98%), Oil red O, and Tris (hydroxymethyl) aminomethane hydrochloride (Tris-HCl) were supplied by Aladdin Reagent Co., Ltd. (Shanghai, China). Sodium dodecyl sulfate (SDS) was provided by Sigma-Aldrich. Span-80 was purchased from Tokyo Chemical Industry Co., Ltd. (Tokyo, Japan). Deionized water was produced from a Milli-Q® water purification system (Merck Millipore, Burlington, MA, USA) and used in all the experiments.
