AMPK agonists Metformin hydrochloride (PHR1084), AICAR (A9978), Resveratrol (R5010); SIRT1 agonist SRT1720 (SRT1720) and NRF-2/HO-1 agonist Protoporphyrin IX cobalt chloride (C1900) were purchased from Sigma-Aldrich. Inhibitors of NF-κB Emodin (E7881) and JSH23 (J4455); IKKβ inhibitor IKK16 (SML1138); STAT1 inhibitor Fludarabine (F2773) and NOS2 inhibitor 1400W (W4262) were purchased from Sigma-Aldrich. Inhibitors of MEK1 and MEK2 PD98059 (ab120234), JNK SP600125 (ab120065) and MAPK SB202190 (ab120638) were purchased from Abcam. Reagents were dissolved in DMSO or water (Metformin and AICAR) at 100x stock concentrations and maximum DMSO concentration used in cells was 1%. Monoclonal antibody against 3-nitrotyrosine (ab61392) was purchased from Abcam. Monoclonal antibody against NF-κB p65 (sc-8008) was purchased from Santa Cruz Biotechnology.

Emodin Modulates LPS-Induced Inflammation

LPS (Escherichia coli, 055: B5) and emodin (E7881, purity ≥90%) were
bought from Sigma (St. Louis, MO, USA). The autophagy inhibitor 3-methyl-3H-purin-6-amine
(3-MA) were the products from Cayman Chemical (Ann Arbor, MI, USA, 13242). The ELISA kits
for judgement of mouse tumor necrosis factor alpha (TNF-α) and IL-6 were generated by
NeoBioscience Technology Co. (Shenzhen, China). Myeloperoxidase (MPO) and lactic acid (LA)
detection kits were bought from Nanjing Jiancheng Bioengineering Institute (Nanjing,
China). Nitric oxide (NO) detect kit was produced by Beyotime Institute of Biotechnology
(Jiangsu, China). Evans Blue was the product from Molbase Institute of Biotechnology
(Shanghai, China). The light Chain 3 (LC3) antibody was bought from Cell Signaling
Technology (Danvers, MA, USA), with BECN1 antibody from Proteintech (Chicago, IL, USA),
GAPDH antibody from Bioss (Bioss, Beijing, China). The BCA protein detect kit,
HRP-combined goat anti-rabbit immune response or defense, and increased chemiluminescence
(ECL) reagents were gained from Thermo Fisher Scientific (Rockford, IL, USA).

Cytotoxicity and Apoptosis Assays

Roswell Park Memorial Institute (RPMI) medium containing phenol red, fetal bovine serum (FBS), and penicillin-streptomycin antibiotics (200 units/ml) were obtained from Gibco BRL (Grand Island, NY, USA). The CellROX™ Green (C10444), CellROX™ Deep Red (C10422), and Hoechst 33342 (H1399) were procured from Thermo Fisher Scientific (USA). The organic solvents were of HPLC-grade (Merck, NJ, USA).
