Cytotoxicity Evaluation of Doxorubicin Formulations

Bovine β-gus (G0376, Type B-3, 2000 units/mg solid) and 4-methylumbelliferyl β-d-glucuronide (M9130, 4-MUG) were purchased from Sigma-Aldrich (Zwijndrecht, The Netherlands). Irgacure 2959 was obtained from Ciba Specialty Chemicals Inc. (Bazel, Switzerland). ABIL EM 90 was provided from Evonik Industries AG (Essen, Germany). All other chemicals and reagents were obtained from Sigma-Aldrich (Zwijndrecht, The Netherlands).

Estrogen and Androgen Receptor Bioassays

Dimethylsulfoxide (DMSO) (minimum 99.7%), hexane (minimum 96%, suitable for CALUX), acetonitrile (minimum 99.99%) and acetone (minimum 99.95%) were purchased from Biosolve (The Netherlands). The estrogen standard 17β-estradiol (minimum 98%), ultra-pure ethanol, Dulbecco’s Modification of Eagle’s Medium high glucose (DMEM without phenol red) and sodium pyruvate (100 mM, sterile-filtered) were obtained from Sigma-Aldrich (Belgium). Minimum Essential Medium Alpha (α-MEM), penicillin-streptomycin, Fetal Bovine Serum (FBS), FBS charcoal-stripped, L-glutamine (200 mM), trypsine, trypsine without phenol red and Phosphate-Buffered Saline (PBS) were obtained from Life Science Technology (United Kingdom). Glass fiber filters MG 227/1/60 with a diameter of 150 mm were purchased from Sartorius (Belgium). Luciferine reagent and lysis reagent were obtained from Promega (The Netherlands). The recombinant, estrogen-responsive BG1Luc4E2 ovarian carcinoma and the androgen-responsive T47D human breast carcinoma cell lines were previously described (Rogers and Denison, 2000 (link); Rogers and Denison, 2002 (link)).

Comparison of Insect Larvae Processing

Hermetia illucens larvae were kindly provided by the Laboratory of Entomology (Wageningen University, The Netherlands). H. illucens larvae were washed and dried before freezing. Tenebrio molitor larvae were purchased from Insectenkwekerij Van de Ven (Deurne, The Netherlands). Alphitobius diaperinus larvae were purchased from Kreca Ento-Feed BV (Ermelo, The Netherlands). Larvae were frozen with liquid nitrogen and stored at −22 °C.
Ultra-high-performance liquid chromatography/mass spectrometry (UHPLC-MS) grade formic acid, methanol, hexane, acetonitrile (ACN) and water were purchased from Biosolve BV (Valkenswaard, The Netherlands). Ethylenediaminetetraacetic acid (EDTA) was purchased from Arcros organics (New Jersey, USA).
All other chemicals used were purchased from Sigma Aldrich (St. Louis, USA) or Merck Millipore (Billerica, USA). Water was acquired using a Milli-Q water purification system (Millipore, Billerica, USA). Ferric iron was used as FeCl3 (purity ≥ 99.0%), unless stated otherwise. Tyrosinase originated from Agaricus bisporus, which was purified to 3000 U/mL as described before31 (link).

Extraction and Analysis of ORURO Chia Seeds

We chose to study the ORURO variety, which is a new genotype bred in France in 2017, adapted to the climatic conditions of the region [34 (link)]. ORURO chia seeds (Salvia hispanica L.) were purchased from the producer Agrofün, Occitania, France.
Purified rapeseed lecithin (Solae Europe SA society, Geneva, Switzerland), rapeseeds (NATUR-Haus Jurgen Kaiser, NK-Wiebelskirchen, Germany) and flax seeds (Linum usitatissimum L.) (Graines de Folie, La E Penell, Normandy, France) were used as for oil extractions (seeds) or NL preparation (rapeseed lecithin).
Ammonium molybdate, internal standard nonadecanoic acid (C19), boron trifluoride diethyl etherate and methanol were purchased from Sigma Aldrich (Saint-Quentin Fallavier, France). Formic acid was obtained from Fluka (Charlotte, NC, USA). Chloroform, hexane, diethyl and petroleum ethers were purchased from Biosolve (Dieuze, France).

LC-MS Analysis of Steroid Hormones

Methanol, hexane and isopropyl-alcohol (IPA) (LC-MS grade) were purchased from Biosolve (Valkenswaard, The Netherlands), CO2 was purchased from Air Liquide (Paris, France). Testosterone (>99%), progesterone (>99%) and -estradiol (>98%) were purchased from Sigma-Aldrich (Overijse, Belgium). The analytes were dissolved at a concentration of 100 µg/mL in pure MeOH. For the measurements where also very low injection volumes were investigated, a concentration of 500 µg/ml was used to obtain high enough UV signals. Some experiments were also performed with a 15/15/70 v%/v%/v% MeOH/IPA/hexane mixture as sample solvent, where the IPA was added to the mixture to allow better mixing of MeOH and hexane [12] . Since the improvement in performance with this sample solvent mixture is in agreement with earlier work [12, 13] and are thus not specific for the feed injector, these results are not discussed in detail. A Zorbax RX-SIL 4.6 x 150 mm with 5 µm particles was used for all experiments (Agilent Technologies, Waldbronn, Germany).

Organic Solvent Extraction Protocol
