Characterization of Seed Nutrients in Chinese Pumpkin Cultivars

Seven commercial cultivars of seed‐rich pumpkin in China, including Dali, Luoren No.2, Luoren No.3, Jinhui No. 4, Ruifeng No.9, Yinhui No. 3, and Xuecheng No.2 (coded LR‐1, LR‐2, LR‐3, JH‐4, RF‐9, YH‐3, and XC‐2), were purchased from Mudanjiang Beidahuang Food Co., Ltd. All seeds were received in good shape (without damage) and were stored at 4℃.
Gamma‐aminobutyric acid (GABA) and phenylisothiocyanate (PITC) were purchased from Sigma Chemical Co. Monosodium glutamate (MSG), trimethylamine, n‐hexane, methyl alcohol, acetonitrile of HPLC grade, and other analytical reagents were obtained from Sinopharm Chemical Reagent Co., Ltd.

Quinoa Bioactive Compound Extraction

Quinoa seed (Mengli-I) was purchased from the Inner Mongolia Yiji Biotechnology Company (Ulanqab, China). Pepsin from porcine gastric mucosa and pancreatin from porcine pancreas were from Sigma-Aldrich (St. Louis, MO, USA). Hydrochloric acid (HCL), sodium hydroxide (NaOH) and n-hexane were acquired from Sinopharm Chemical Reagent Co. Ltd. (Shanghai, China). The VivaFlow 200 tangential flow ultrafiltration system with a molecular weight cut-off (MWCO) of 5 kDa (Sartorius Stedim Biotech, Goettingen, Germany) was used in the ultrafiltration. The primers were synthesized by Sangon Biotech Corporation (Shanghai, China) and the primers used in this work are shown in Table S1. Specific primary antibodies against GAPDH, HDAC1, NFκB, IL-6, IL-8, Bcl-2, caspase3 were purchased from Bioss Inc. (Beijing, China).

Cracking of LDPE via Zeolites

1,4-Cyclohexanedimethanol, 1,5-pentanediol, 2,2-dimethoxypropane, and p-toluene sulfonic acid (PTSA) were purchased from the Aladdin Reagent Company (Shanghai, China). Low-density polyethylene (LDPE) powder used as a model feedstock in the cracking reaction was provided by Alfa Aesar (Stock #42607) (Tianjin, China).

Enzymatic Enrichment of Fish Oil

Two types of fish oils, namely triglycerides fish oil (19.21% EPA and 10.81% DHA) and ethyl ester fish oil (42.47% EPA and 31.91% DHA), were kindly provided by the Fujian Coland Marine Bio‐engineering Co., Ltd (Fujian, China). Ionic liquids, namely 1‐butyl‐3‐methylimidazolium tetrafluoroborate ([BMIM][BF4]), 1‐butyl‐3‐methylimidazolium hexafluorophosphate ([BMIM][PF6]), and 1‐butyl‐3‐methylimidazolium bis(trifluoromethanesulfonyl)imide ([BMIM][Tf2N]), were purchased from Shanghai Chengjie Chemical Co., Ltd (Shanghai, China). N‐octanoic acid and n‐hexane (HPLC grade) were purchased, respectively, from Sinopharm Chemical Reagent Co., Ltd. (Shanghai, China) and Shandong Yuwang industrial Co., Ltd (Shandong, China). Thin‐layer chromatography silica gel G (analytical grade) was purchased from Qingdao Marine Chemical Co., Ltd (Shandong, China). Boron trifluoride diethyl ether and carboxymethyl cellulose sodium (analytical grade) were purchased from Aladdin Chemicals Co., Ltd (Shanghai, China).

A TG16-WS high-speed centrifuge was used from Changsha Xiangyi Centrifuge Instrument Co., Ltd. (Changsha, China). A SB-3200 ultrasonic cleaner was used from Kunshan Ultrasonic Instrument Co., Ltd. (Shanghai, China). A DHG-9070A electrothermal oven thermostat blast was used from the Shanghai Yiheng Science and Technology Co., Ltd. (Shanghai, China).

Phytase Extraction from Transgenic Soybean

Mature seeds from the mAppA transgenic line were pulverized to permit passage through a 0.38 mm sieve36 . The extraction mixture consisted of 1 g of collected soybean flour42 (link) and 5 mL of n-hexane (97%, Sinopharm Chemical Reagent Co., Ltd). The ratio of solvent to beans was 5:143 (link). Experiments were performed at 55, 60 or 65 °C44 (link),45 (link), with extraction time limit set to 1, 2 or 3 hs at each temperature point. All the extractions were carried out in triplicate. At the end of the contact time, the supernatants were discarded by centrifugation (15938 g, 10 min) and traces of solvent remaining were removed by evaporation. The resulting precipitation was homogenized in 10 mL of 0.2 M sodium acetate buffer (pH 5.0) for phytase extraction. The phytase in the control sample (1 g of soybean flour without n-hexane extraction) was extracted simultaneously with 10 mL of 0.2 M sodium acetate buffer (pH 5.0). After centrifugation (15938 g, 10 min), the supernatants were collected and assayed for phytase activity using the standard method. The phytase activity per gram for both non n-hexane-extracted and n-hexane extracted samples were calculated based on original pulverized soybean weight.
