Antibodies against GAPDH (60004-1-Ig), α-tubulin (66031-1-Ig), Flag (DDDDK tag) (20543-1-AP), SOCS3 (14025-1-AP), p21 (10355-1-AP), and cyclin D1 (60186-1-Ig) were purchased from Proteintech. Anti-ELK1 antibody (ab32106) was purchased from Abcam (Shanghai, China).

Isolation and Culture of Human Amniotic Epithelial Cells

For the isolation and culture of hAECs, the amnion was mechanically peeled from the underlying chorion and washed three times with phosphate-buffered saline (E607008, PBS, Sangon Biotech, China) supplemented with 10% penicillin and streptomycin (15070063, Thermo Fisher Scientific, USA). The amnion was incubated with 0.25% trypsin-EDTA (25200072, Thermo Fisher Scientific, USA) for 15 min at 37°C, and the human amniotic epithelial cells were mechanically detached from the amnion. Dispersed hAECs were collected via centrifugation at 1500 rpm for 10 min, plated in 10-cm cell culture dishes in DMEM/F-12 medium (11330032, Thermo Fisher Scientific, USA) supplemented with 10% fetal bovine serum (FBS, ExCell Bio, China), 10 ng/mL human recombinant epidermal growth factor (EGF, PHG0311, Thermo Fisher Scientific, USA), 5 μM SB431542 (301836-41-9, Selleck, China) and 1% penicillin and streptomycin, and cultured in a cell incubator at 37°C. The hAEC culture medium was changed every 2 days.
Human corneal epithelial cells (hCECs) and human corneal stromal cells (hCSCs) were a kind gift of professor Qingjun Zhou (Shandong Eye Institute, China) and cultured in DMEM/F12 medium supplemented with 10% FBS in a cell incubator at 37°C.

Culturing Breast Cancer Cell Lines

The breast cancer cell line MCF7 was cultured in DMEM/high glucose (SH30243.01B, HyClone, Logan, UT) with 10% fetal bovine serum (FBS) (04-001-1ACS, Biological Industries, Kibbutz Beth Haemek, Israel) and 1% penicillin/streptomycin (p/s) (C0222, Beyotime Institute of Biotechnology, Jiangsu, China). SKBR3, HCC1806 and 786-0 cells were cultured in RPMI-1640 (SH30809.01B, HyClone) containing 10% FBS and 1% p/s. MDA-MB-231 cells were cultured in DMEM/F12 (SH30023.01B, HyClone), supplemented with 10% FBS and 1% p/s. The immortalized human breast epithelial cell line MCF10A was maintained in DMEM/F12, supplemented with 5% horse serum (16050-130, Gibco, New Zealand), 20 ng/mL EGF (PHG0311, Invitrogen, Carlsbad, CA), 0.5 mg/mL hydrocortisone (MB1567, Meilunbio, Dalian, China), 100 ng/mL cholera toxin (C8052, Sigma-Aldrich, St. Louis, MO), 10 μg/mL insulin (Wanbang Biopharmaceuticals, Xuzhou, China), and 1% p/s. The cells were purchased from Conservation Genetics CAS Kunming Cell Bank. Cell lines were tested to be mycoplasma-free by PCR (16 (link)).

Vitamin E (25985), hygromycin B (hydrate: 14291), reparixin (21492), and atorvastatin (10493) were purchased from Cayman (Cayman). atorvastatin (Lipitor; VITARIS) was used for feeding mice.
